Summary. A case of atypical polyagglutinability due to the Tn antigen is described. The patient, although apparently healthy, has leucopenia and thrombocytopenia. Her cells presented the usual difficulties of polyagglutinable cells due to the presence of anti‐Tn in typing reagents whether of human or animal origin.

A murine monoclonal antibody, MLS 128, that was assigned to an anti-Tn antibody has been established by immunizing mice with human colonic cancer cells (LS 180). MLS 128 bound to mucin glycopeptides from LS 180 cells and their asialo forms to the same extent as well as to ovine submaxillary mucin (OSM) and asialo OSM.

Enzymatic synthesis of the Tn antigen

The enzymatic synthesis of the Tn antigen (GalNAc--O-Ser), a glyco-aminoacid of great biological importance, is reported. The reaction was promoted by commercial -N-acetylgalactosaminidase from Acremonium sp., using p-nitrophenyl--N-acetylgalactosamine as the donor. The kinetics were monitored by capillary electrophoresis and LC–UV-MS.
