Combination of tocolytic agents for inhibiting preterm labour [PDF]
Preterm birth represents the single largest cause of mortality and morbidity for newborns and a major cause of morbidity for pregnant women. Tocolytic agents include a wide range of drugs that can inhibit labour to prolong pregnancy. This may gain time to allow the fetus to mature further before being born, permit antenatal corticosteroid ...

Effects of sympathomimetic tocolytic agents on the fetus [PDF]
The variety of factors controlling the onset of parturition, as well as the very complex mechanisms which physiologically maintain pregnancy by inhibiting uterine contractions are only partially understood today. As long as the triggering mechanisms, and the conditions favoring timely and early labor are incompletely known, symptomatic therapeutic ...

School Performance After Maintenance Tocolysis With Nifedipine for Threatened Preterm Birth: 12-Year Follow-Up of the APOSTEL 2 Trial. [PDF]
ABSTRACT Objective To evaluate school performance at 12 years of age in children prenatally exposed to maintenance tocolysis with nifedipine versus placebo. Design 12‐year follow‐up of the multicentre APOSTEL 2 trial, in which participants with threatened preterm birth between 26+0 and 32+2 weeks of gestation, who remained pregnant after initial 48‐h ...

The oxytocin-oxytocin receptor system and its antagonists as tocolytic agents. [PDF]
Oxytocin, a hormone involved in numerous physiologic processes, plays a central role in the mechanisms of parturition and lactation. It acts through its receptor, which belongs to the G-protein-coupled receptor superfamily, while Gq/phospholipase C (PLC)/inositol 1,4,5-triphosphate (InsP3) is the main pathway via which it exerts its action in the ...

Tocolytic agents and the failure to improve neonatal outcomes
Abstract Premature delivery has long been a major problem in obstetrics. Increased perinatal morbidity and mortality as well as substantial health care costs, both short and long term, have driven the efforts to combat this complication. While multiple agents have been studied over the last 70 years leading to hundreds of publications

Efficacy of oral nifedipine as a tocolytic agent
Background: Preterm birth is defined as birth at less than 37 weeks period of gestation, is the most important single determinant of adverse infant outcome in terms of both survival and quality of life. The need for tocolysis in terms of safety and efficacy is necessary to decrease perinatal mortality and morbidity in preterm labour.

This study shows that tafoxiparin 300 mg can increase the rate of cervical ripening and the likelihood of spontaneous onset of labor, while also reducing the duration of labor in women achieving spontaneous onset of labor. The favorable safety profile of tafoxiparin suggests suitability for routine clinical use, including home administration.

Therapeutic role of nifedipine in threatened preterm labor: Current evidence and future perspectives
Abstract Preterm birth occurs in approximately 10% of all pregnancies, and is not only the leading cause of neonatal mortality but also a major contributor to short‐ and long‐term morbidities due to immaturity. Preterm birth has also been linked to an increased risk of maternal cardiovascular and cerebrovascular diseases, making it a critical concern ...
