Targeted Anti‐IL‐1 Immunomodulatory Therapy in Pediatric Onset PPP1R13L‐Related Arrhythmogenic Cardiomyopathy

open access: yesAmerican Journal of Medical Genetics Part A, EarlyView.
ABSTRACT Autosomal recessive loss‐of‐function variants in PPP1R13L cause an ultra‐rare cardiocutaneous syndrome characterized by rapidly progressive arrhythmogenic cardiomyopathy (ACM). PPP1R13L encodes iASPP, which has two potentially overlapping mechanisms driving ACM as both a regulator of NFκB‐mediated inflammation and a binding partner within the ...

Transfersomes with core and surface‐loaded NF‐κB p65 siRNA for enhanced transdermal transfection and effective treatment of psoriasis

open access: yesBMEMat, EarlyView.
TCPL, a transfersome with Tween 80 and PEI, enables efficient transdermal siNF‐κB delivery, resolving PEI's toxicity issue. It promotes lysosomal escape and effective transfection, silences NF‐κB, modulates immunity, and inhibits angiogenesis, alleviating psoriasis. With good biocompatibility, TCPL holds strong clinical translation potential. Schematic

Impaired RelA signaling and lipid metabolism dysregulation in hepatocytes: driving forces in the progression of metabolic dysfunction-associated steatotic liver disease

open access: yesCell Death Discovery
RelA, also known as nuclear factor kappa B p65, plays a crucial role in the pathogenesis of various liver diseases. However, the specific role of RelA in hepatocytes during the progression of metabolic dysfunction-associated steatotic liver disease ...

NF-κB regulates DNA double-strand break repair in conjunction with BRCA1-CtIP complexes [PDF]

open access: yes, 2011
NF-κB is involved in immune responses, inflammation, oncogenesis, cell proliferation and apoptosis. Even though NF-κB can be activated by DNA damage via Ataxia telangiectasia-mutated (ATM) signalling, little was known about an involvement in DNA repair ...

Immunomodulation of the Prostate Tumor Microenvironment Following Inorganic Arsenic Exposure

open access: yesJournal of Applied Toxicology, EarlyView.
ABSTRACT The tumor microenvironment (TME) influences prostate cancer (PCa) progression through stromal and immune interactions. Adipose‐derived mesenchymal stromal cells (ASCs) modulate immune tone, while inorganic arsenic (iAs), a widespread toxicant, is linked to immune suppression and carcinogenesis.

Feedback loop between hepatocyte nuclear factor 1α and endoplasmic reticulum stress mitigates liver injury by downregulating hepatocyte apoptosis

open access: yesScientific Reports, 2022
Hepatocyte nuclear factor alpha (HNF1α), endoplasmic reticulum (ER) stress, and hepatocyte apoptosis contribute to severe acute exacerbation (SAE) of liver injury. Here, we explore HNF1α–ER stress-hepatocyte apoptosis interaction in liver injury.

Transcriptional Induction of ADAMTS5 Protein by Nuclear Factor-κB (NF-κB) Family Member RelA/p65 in Chondrocytes during Osteoarthritis Development [PDF]

open access: yesJournal of Biological Chemistry, 2013
Here we sought to identify transcription factors that induce ADAMTS5, a crucial proteinase for osteoarthritis development. Exhaustive comparison of the genomic sequences of human, macaque, and mouse ADAMTS5 genes revealed that the proximal 1.4 kb of the 5'-end-flanking regions containing several consensus motifs was highly conserved.

Immunomodulatory and Cytotoxic Properties of Enniatin B1 in Porcine Alveolar Macrophages

open access: yesJournal of Applied Toxicology, EarlyView.
ABSTRACT This study investigates the immunomodulatory effects of Enniatin B1 (ENNB1), an emerging mycotoxin, on porcine alveolar macrophages (PAMs), a species‐specific model relevant to human innate immunity. PAMs were exposed to increasing ENNB1 concentrations (0.5–6 μM) over 6, 24, and 48 h.

Investigation of the Mechanism of Cinnamaldehyde in Irritable Bowel Syndrome Based via Network Pharmacology, Molecular Docking, and Animal Experiments

open access: yesPediatric Discovery, EarlyView.
Through network pharmacology and molecular docking, it has been discovered that CA can target MAOB, among other proteins, to exert a therapeutic effect in IBS. In vivo, CA lowered visceral hypersensitivity, anxiety and depression‐like behaviors, and fecal water content, highlighting its therapeutic potential for IBS via anti‐inflammatory pathways ...
