Functional immunoglobulin transgenes guide ordered B-cell differentiation in Rag-1-deficient mice [PDF]

open access: yes, 1994
We have examined the regulatory role of the individual components of the immunoglobulin antigen receptor in B-cell development by transgenic complementation of Rag-1 deficient (Rag-1⁻) mice. Complementation with a membrane µ heavy chain (µHC) gene allows

The role of the Cucumber mosaic virus 2b protein in viral movement and symptom induction [PDF]

open access: yes, 2009
The Cucumber mosaic virus (CMV) 2b protein is a counter-defense factor and symptom determinant. Conserved domains in the 2b protein sequence were mutated in the 2b gene of strain Fny-CMV.

Frequent Occurrence of Transgene Deletion in Transgenic Plants

open access: yesMolecules and Cells, 1998
The status of the transgene in tobacco plants transformed by Agrobacterium was analyzed with PCR. Twelve percent of the transgenic plants with the nptII gene showed different levels of transgene deletion, which was also found in transgenic watermelon (10-30%) and carrot (40-60%).

T Cell Exhaustion in Cancer Immunotherapy: Heterogeneity, Mechanisms, and Therapeutic Opportunities

open access: yesAdvanced Science, EarlyView.
T cell exhaustion limits immunotherapy efficacy. This article delineates its progression from stem‐like to terminally exhausted states, governed by persistent antigen, transcription factors, epigenetics, and metabolism. It maps the exhaustion landscape in the TME and proposes integrated reversal strategies, providing a translational roadmap to overcome

Transcriptome Analysis of Targeted Mouse Mutations Reveals the Topography of Local Changes in Gene Expression. [PDF]

open access: yes, 2016
The unintended consequences of gene targeting in mouse models have not been thoroughly studied and a more systematic analysis is needed to understand the frequency and characteristics of off-target effects.

Postsynaptic Complexin Mediates Constitutive Exocytosis of Nicotinic Acetylcholine Receptor

open access: yesAdvanced Science, EarlyView.
Beyond its established presynaptic role, complexin is identified as a postsynaptic regulator of neurotransmitter receptor trafficking. By promoting constitutive and activity‐dependent exocytosis of nicotinic acetylcholine receptors, complexin controls spontaneous synaptic signaling at the C.

Chimeric piggyBac transposases for genomic targeting in human cells. [PDF]

open access: yes, 2012
Integrating vectors such as viruses and transposons insert transgenes semi-randomly and can potentially disrupt or deregulate genes. For these techniques to be of therapeutic value, a method for controlling the precise location of insertion is required ...

Cis‐ and Trans‐Regulatory Factors Independently Shape Phenotypic Heterogeneity of Retinitis Pigmentosa

open access: yesAdvanced Science, EarlyView.
A zebrafish model carrying an identical human RHO S334X allele reveals two independent genetic layers shaping retinitis pigmentosa (RP) severity: a protective 3‐bp cis‐regulatory insertion that attenuates transgene expression, and a dominant trans‐acting modifier that restores a severe phenotype.

Extended treatment with MY-NEOVAX, personalized neoantigen-enhanced oncolytic viruses, for two end-stage cancer patients. [PDF]

open access: yes, 2019
Neoantigen vaccines involving multi-peptides and poly-epitope-encoding RNA or DNA have undergone early phase clinical testing with modest reported antitumor effects [ 1].

Transgenic Animal Technology

open access: yesJournal of Andrology, 1997
There are numerous tools available to modify the genetic makeup of animals. They are being used to good advantage for studying basic biological phenomena. Within the decade, biomedical products derived from transgenic animals will be available, but the use of this technology for enhancing the quality and efficiency of livestock production will await ...
