Remote Magnetomechanical Neuromodulation Uncovers Therapeutic Mechanisms for Alleviating Parkinsonian Symptoms in Freely Moving Mice

open access: yesAdvanced Science, EarlyView.
Magnetomechanical neuromodulation using magnetic nanodiscs enables remote activation of neurons. In a hemiparkinsonian mouse model, alternating magnetic fields actuate the nanodiscs to generate torque that opens mechanosensitive ion channels within the subthalamic nucleus, thereby modulating basal ganglia motor circuitry.

RDR2 partially antagonizes the production of RDR6-dependent siRNA in sense transgene-mediated PTGS.

open access: yesPLoS ONE, 2012
BackgroundRNA-DEPENDENT RNA POLYMERASE6 (RDR6) and SUPPRESSOR of GENE SILENCING 3 (SGS3) are required for DNA methylation and post-transcriptional gene silencing (PTGS) mediated by 21-nt siRNAs produced by sense transgenes (S-PTGS).

Modified genomic transgenes to assess individual miR-279/996 functions.

open access: yes, 2015
(A) In the 16.6kb mir-279/996 rescue transgene, the 5' end extends to cover a portion of the upstream CG14508 gene and 3' end extends into the downstream Ef1gamma gene. Green and blue triangles represent mir-279 and mir-996 hairpins, respectively.

Targeting Endogenous Lipophagy: A Novel Strategy to Enhance MSC Osteogenesis and Mineralization for Senile Osteoporosis Therapy

open access: yesAdvanced Science, EarlyView.
Schematic representation of the role of lipophagy in bone mesenchymal stem cells(MSCs). In healthy MSCs, functional lipophagy efficiently degrades lipid droplets to support oxidative phosphorylation and cellular energy production, thereby facilitating osteogenic differentiation and matrix mineralization.

Transgenic Animal Technology

open access: yesJournal of Andrology, 1997
There are numerous tools available to modify the genetic makeup of animals. They are being used to good advantage for studying basic biological phenomena. Within the decade, biomedical products derived from transgenic animals will be available, but the use of this technology for enhancing the quality and efficiency of livestock production will await ...

Rescue Expression of dSarm1 transgenes rescue lethality of dSarm1 mutants.

open access: yes, 2022
Daughterless-GAL4 (da-GAL4) was used to drive ubiquitous expression of Sarm1 transgenes. Homozygous dSarm14705/dSarm14621 mutant animals are lethal. Expression of transgenes in the mutant background tests for the ability of the transgene to rescue dSarm1

Boosting Sensory Nerve‐to‐Bone Interactions Enhances Hedgehog Mediated Calvarial Bone Repair

open access: yesAdvanced Science, EarlyView.
Boosting sensory nerve activity via TrkA agonism strongly accelerates calvarial bone repair in adult mice. Furthermore, single‐cell RNA sequencing and neuron–bone interactome analyses identify these sensory neurons as a direct neural source of Hedgehog pathway ligands. Consequently, these ligands drive osteoblast differentiation of skeletal progenitors,

Scalable Engineering of Bio‐Manufactured Extracellular Vesicles for Selective Delivery in Ovarian Cancer Patient‐Derived Models

open access: yesAdvanced Science, EarlyView.
Engineered extracellular vesicles displaying Ephrin‐B2 selectively target Ephrin‐B4–expressing ovarian cancer cells, enabling precise delivery in patient‐derived models. This scalable bio‐manufacturing platform reveals a versatile strategy to exploit Ephrin signaling for highly specific therapeutic payload delivery and motivates exploration of tailored

Stable transgenes bear fruit

open access: yes, 2008
Analysis of the transgenic papaya genome sequence suggests that transgenes generally stay put following integration and can achieve stable expression level from generation to generation.
