Construction of an Oral Vaccine for Transmissible Gastroenteritis Virus Based on the TGEV N Gene Expressed in an Attenuated Salmonella Typhimurium Vector

Journal of Virological Methods, 2016
This research aimed to develop an oral vaccine for transmissible gastroenteritis virus (TGEV) based on the TGEV N gene expressed in an attenuated Salmonella typhimurium vector and aimed to evaluate the vaccine's immune response in piglets. Recombinant plasmid pVAX-N was transformed into competent cells of attenuated S.

Inhibitory effects of recombinant porcine interferon-α on porcine transmissible gastroenteritis virus infections in TGEV-seronegative piglets

Veterinary Microbiology, 2021
Our previous research obtained purified recombinant porcine interferon-α (rPoIFN-α) containing thioredoxin (Trx) fusion tag in E. coli Rosetta (DE3). Here, we evaluate the efficacy of this rPoIFN-α to prevent piglets from the infection of the transmissible gastroenteritis virus (TGEV) attack.

In Situ Hybridization Technique for the Detection of Swine Enteric and Respiratory Coronaviruses, Transmissible Gastroenteritis Virus (TGEV) and Porcine Respiratory Coronavirus (PRCV), in Formalin-Fixed Paraffin-Embedded Tissues

open access: yesJournal of Virological Methods, 1996
The in situ hybridization (ISH) technique was developed to detect the swine coronaviruses, transmissible gastroenteritis virus (TGEV) and porcine respiratory coronavirus (PRCV), in cell culture and tissue sections from TGEV-or PRCV-infected pigs. The 35S-labeled RNA probes were generated from two plasmids pPSP.FP1 and pPSP.FP2 containing part of the S ...

The Genome of Transmissible Gastroenteritis Virus (TGEV)

1984
Lysis of isolated TGE virions with sodium dodecyl sulphate (SDS) at room temperature followed by electrophoresis, yields predominantly a single band of RNA running at about molecular weight 6.8 × 106. Heating the SDS lysate at 80°C for 15 min prior to electrophoresis results in a cluster of bands of between 0.2–6.8 × 106.

Antigenic variation among transmissible gastroenteritis virus (tgev) and porcine respiratory coronavirus strains detected with monoclonal antibodies to the S protein of tgev

American Journal of Veterinary Research, 1992
Summary Five nonneutralizing monoclonal antibodies (MAb) generated to the virulent Miller strain of transmissible gastroenteritis virus (tgev) and specific for the S protein were characterized. Competition assays between purified and biotinylated MAb indicated that MAb 75B10 and 8G11 mapped near a new subsite, designated V and 2 MAb, 44C11 and 45A8 ...

Active and passive immune responses to transmissible gastroenteritis virus (TGEV) in swine inoculated with recombinant baculovirus-expressed TGEV spike glycoprotein vaccines

American Journal of Veterinary Research, 1997
Abstract Objective Baculovirus-expressed transmissible gastroenteritis virus (TGEV) spike (S) glycoprotein vaccines were inoculated parenterally in swine to determine whether such vaccines could induce serum and whey virus-neutralizing (VN) antibodies and protective lactogenic immunity for TGEV-challenge-exposed pigs.

Characterization of the Transmissible Gastroenteritis Virus (TGEV) Transcription Initiation Sequence

1995
The ability of the TGEV transcription initiation sequence (TIS) to produce subgenomic RNAs was investigated by placing a reporter gene, chloramphenicol acetyltransferase (CAT) under the control of either the mRNA 6 or the mRNA 7 TISs. Both constructs only produced CAT in TGEV infected cells and the amount of CAT produced from the mRNA 7 TIS was less ...

Intragastric Administration of Attenuated Salmonella Typhimurium Harbouring Transmissible Gastroenteritis Virus (TGEV) DNA Vaccine Induced Specific Antibody Production

open access: yesVaccine, 2009
Attenuated Salmonella typhimurium was selected as a transgenic vehicle for the development of live mucosal vaccines against transmissible gastroenteritis virus (TGEV). A 2.2kb DNA fragment, encoding for N-terminal domain glycoprotein S of TGEV, was amplified by RT-PCR and cloned into eukaryotic expression vector pVAX1.

Immune response of sows vaccinated with attenuated transmissible gastroenteritis virus (TGEV) and recombinant TGEV spike protein vaccines and protection of their suckling pigs against virulent TGEV challenge exposure

American Journal of Veterinary Research, 1998
Abstract Objective To compare recombinant transmissible gastroenteritis virus (TGEV) spike protein, (SP) R2-2, with attenuated live virus (ALV) vaccine in sows during late pregnancy. Animals 13 TGEV-seronegative sows and their pigs. Procedure At prepartum weeks (PPW) 6 and 4, sows of groups 1 and 2 received ALV via the oral/intranasal (O/IN) route.

Competition ELISA, using monoclonal antibodies to the transmissible gastroenteritis virus (TGEV) S protein, for serologic differentiation of pigs infected with TGEV or porcine respiratory coronavirus

American Journal of Veterinary Research, 1993
SUMMARY Monoclonal antibodies (MAB) to subsite A (25C9) and subsite D (44C11) of the S protein of transmissible gastroenteritis virus (tgev) were used in a blocking elisa on fixed tgev-infected swine testis cells to differentiate sera from pigs experimentally inoculated with either tgev or porcine respiratory coronavirus (prcv).
