A Living Multi‐Omics PDX Biobank of High‐Risk Sarcomas Reveals ecDNA‐Driven Oncogene Amplification in Osteosarcoma and Enables Preclinical Precision Oncology

open access: yesAdvanced Science, EarlyView.
A living multi‐omics sarcoma PDX biobank is enriched for high‐risk disease and preserves key patient tumor features at histopathological and molecular levels. Integrated profiling reveals ecDNA‐driven oncogene amplification in osteosarcoma and preferential retention of ecDNA during PDX evolution.

Aged ZnO Nanoparticles Promote Breast Cancer Lung Metastasis by Remodeling the Pulmonary Microenvironment Through the CXCL10/Wnt/β‐Catenin Axis

open access: yesAdvanced Science, EarlyView.
In summary, pulmonary exposure to ZnO NPs, particularly aged NPs, promotes breast cancer metastasis by remodelling the lung tumour microenvironment. Aged ZnO NPs enrich M2‐macrophages, MDSCs and activated CAFs to form an immunosuppressive metastatic niche. CXCL10 from these cells drives metastasis via Wnt/β‐catenin activation.

Serial H&E Pathology Reveals Progression‐Associated Nuclear Size Drift as a Longitudinal Digital Readout of Bladder Cancer Evolution

open access: yesAdvanced Science, EarlyView.
Routine serial H&E specimens from bladder cancer are converted into paired T0/T1 digital pathology readouts of tumor evolution. Progression is associated with longitudinal nuclear drift, with nuclear size‐related changes most evident in the independent confirmation cohort, alongside a broader spatial burden of increased nuclear area and biological ...

Redefining the Role of Splenectomy in Scirrhous Gastric Cancer: Toward a Selective Multimodal Strategy

open access: yesAnnals of Gastroenterological Surgery, EarlyView.
ABSTRACT Scirrhous gastric cancer, characterized by diffuse infiltration and a high propensity for peritoneal dissemination, remains one of the most aggressive subtypes of gastric cancer. Despite a relatively high incidence of splenic hilar lymph node (No. 10) metastasis, the oncologic benefit of splenectomy remains controversial, while its association

Can Chemoradiotherapy Expand Immune Checkpoint Blockade Responsiveness in Mismatch Repair‐Proficient Rectal Cancer?

open access: yesAnnals of Gastroenterological Surgery, EarlyView.
ABSTRACT Immune checkpoint blockade (ICB) has transformed the treatment of mismatch repair‐deficient (dMMR) colorectal cancer but produces no meaningful benefit in advanced mismatch repair‐proficient (pMMR) disease. In the neoadjuvant setting, however, pathological responses have been reported in a subset of pMMR tumors, suggesting that clinical ...

Composition‐Aware Cross‐Sectional Integration for Spatial Transcriptomics

open access: yesAdvanced Intelligent Discovery, EarlyView.
Multi‐section spatial transcriptomics demands coherent cell‐type deconvolution, domain detection, and batch correction, yet existing pipelines treat these tasks separately. FUSION unifies them within a composition‐aware latent framework, modeling reads as cell‐type–specific topics and clustering in embedding space.

Spatially Informed Feature Selection and Machine Learning in Matrix‐Assisted Laser Desorption/Ionization Imaging for Cohort‐Scale Molecular Tissue Phenomics in Glioblastoma

open access: yesAdvanced Intelligent Discovery, EarlyView.
Matrix‐assisted laser desorption/ionization imaging‐based identification of reliable small molecule markers across heterogeneous glioblastoma cohorts is challenging with intensity‐only methods. We present spatially informed feature selection (SIFS), a spatially informed framework that prioritizes molecules consistently colocalizing with histopathology.
