Phosphatidylinositol-3,5-bisphosphate lipid-binding-induced activation of the human two-pore channel 2 [PDF]

open access: yesCellular and Molecular Life Sciences, 2018
Mammalian two-pore channels (TPCs) are activated by the low-abundance membrane lipid phosphatidyl-(3,5)-bisphosphate (PI(3,5)P2) present in the endo-lysosomal system. Malfunction of human TPC1 or TPC2 (hTPC) results in severe organellar storage diseases and membrane trafficking defects.

Two‐pore channel 2 (TPC2) Role in Melanoma

The FASEB Journal, 2020
Two‐pore channels (TPC), a recently described class of NAADP‐ and PI(3,5)P2–sensitive calcium‐permeable cation channels in the endolysosomal system of cells have a crucial rule in cellular functions and have been linked to various type of diseases including cancer.

Neighbourhood Watch: Two-pore-2 channels talking to IP3 receptors

Cell Calcium
The recent elegant study by Y. Yuan and colleagues examined functional relationships between the lysosomal two-pore channels 2 (TPC2) and IP3 receptors (IP3Rs) located in the endoplasmic reticulum [1]. The findings of this study suggest functional coupling of these channels and receptors.

Determinants of pH sensing in the two-pore domain K+ channels TASK-1 and -2

Pflügers Archiv - European Journal of Physiology, 2003
TASK-1 and -2 are members of the two-pore domain potassium (K(+)) channel family and are sensitive to changes in extracellular pH. The effects of mutating charged, extracellular-facing residues in TASK-1 and -2 were studied in Xenopusoocytes by two-electrode voltage clamp. Hydrogen ion block was independent of voltage with K(d) values of 149+/-17.9 nM [

Interrupting two-pore channels 2 (TPC2) and sperm function enhancement

2023
The proper functioning of sperm is essential for successful fertilization. Various physiological regulators control sperm motility, energy balance, capacitation, and the acrosomal reaction. Intracellular calcium ion (Ca2+) concentration and pH levels play a crucial role in regulating sperm physiology.
