Design and construction of a structural model of the bifunctional GlmU protein in complex with N‐acetyl‐D‐glucosamine‐1‐phosphate and uridine‐diphosphate‐N‐acetylglucosamine

open access: yesThe FASEB Journal, 2018
The focus of this project was to explore the structure of the GlmU protein from E. coli (PDB ID: 2OI7) and to design and build a physical model that illustrates the key functional features of the protein.

Sequence of a cDNA that specifies the uridine diphosphate N-acetyl-D-glucosamine:dolichol phosphate N-acetylglucosamine-1-phosphate transferase from Chinese hamster ovary cells

open access: yesJournal of Biological Chemistry, 1990
We have isolated a portion of the uridine diphosphate N-acetyl-D-glucosamine:dolichol phosphate N-acetyl-glucosamine-1-phosphate transferase gene (GTR2) from the genome of a tunicamycin-resistant clonal Chinese hamster ovary cell line, 3E11. The genomic fragment was selected by its hybridization to the yeast ALG-7 gene at low stringency.

Characterization of GDP-mannose Pyrophosphorylase from Escherichia Coli O157:H7 EDL933 and Its Broad Substrate Specificity

open access: yes, 2005
GDP-mannose pyrophosphorylase gene (ManC) of Escherichia coli (E. coli) O157 was cloned and expressed as a highly soluble protein in E. coli BL21 (DE3). The enzyme was subsequently purified using hydrophobic and ion exchange chromatographies. ManC showed

Aberrant O-GlcNAcylated proteins: New perspectives in breast and colorectal cancer

open access: yesFrontiers in Endocrinology, 2014
Increasing glucose consumption is thought to provide an evolutionary advantage to cancer cells. Alteration of glucose metabolism in cancer influences various important metabolic pathways including the hexosamine biosynthesis pathway (HBP), a relatively ...

The kinase Isr1 negatively regulates hexosamine biosynthesis in S. cerevisiae.

open access: yesPLoS Genetics, 2020
The S. cerevisiae ISR1 gene encodes a putative kinase with no ascribed function. Here, we show that Isr1 acts as a negative regulator of the highly-conserved hexosamine biosynthesis pathway (HBP), which converts glucose into uridine diphosphate N ...

Bacterial lipid II analogs : novel in vitro substrates for mammalian oligosaccharyl diphosphodolichol diphosphatase (DLODP) activities [PDF]

open access: yes, 2019
Mammalian protein N-glycosylation requires the transfer of an oligosaccharide containing 2 residues of N-acetylglucosamine, 9 residues of mannose and 3 residues of glucose (Glc3Man9 GlcNAc2) from Glc3Man9GlcNAc2-diphospho (PP)-dolichol (DLO) onto ...

O-GlcNAc transferase in astrocytes modulates depression-related stress susceptibility through glutamatergic synaptic transmission

open access: yesThe Journal of Clinical Investigation, 2023
Major depressive disorder is a common and devastating psychiatric disease, and the prevalence and burden are substantially increasing worldwide. Multiple studies of depression patients have implicated glucose metabolic dysfunction in the pathophysiology ...

Clinical, biochemical and molecular characterization of Korean patients with mucolipidosis II/III and successful prenatal diagnosis [PDF]

open access: yes, 2017
Background: Mucolipidosis types II and III (ML II/III) are autosomal recessive disorders caused by a deficiency in the lysosomal enzyme N-acetylglucosamine-1-phosphotransferase.

Corynebacterium glutamicum for Microbial Production of Chitin Oligosaccharides Using Modular Engineering

open access: yesFood Bioengineering
Microbial fermentation is a potent strategy for eco‐friendly and sustainable chitin oligosaccharide (CHOS) production. Nonetheless, hurdles (e.g., imbalanced metabolic flow and the need for uridine diphosphate (UDP)‐sugar donor consumption in CHOS ...

Recent advances in small molecule LpxC inhibitors against gram-negative bacteria (2014–2024)

open access: yesFrontiers in Microbiology
In 2024, WHO added multiple multidrug-resistant (MDR) Gram-negative bacteria to the bacteria priority pathogens list, and the continued increase in MDR Gram-negative bacteria poses a serious threat to public health.
