Synthetic biology-inspired development of live attenuated influenza vaccines

open access: yesnpj Vaccines
Live attenuated influenza vaccines (LAIVs) provide robust, cross-protective immunity but have traditionally been developed empirically and are associated with safety concerns.

Histo-blood group antigens as receptors for rotavirus, new understanding on rotavirus epidemiology and vaccine strategyRotavirus host receptor and vaccine strategy

open access: yesEmerging Microbes and Infections, 2017
The success of the two rotavirus (RV) vaccines (Rotarix and RotaTeq) in many countries endorses a live attenuated vaccine approach against RVs. However, the lower efficacies of both vaccines in many low- and middle-income countries indicate a need to ...

Secreted Nonstructural Protein 3 is a Pathogenic Determinant of Orbivirus

open access: yesAdvanced Science, EarlyView.
This study uncovers a conserved PIP2‐dependent secretory pathway of orbivirus NS3 that induces vascular leakage. Pharmacological disruption of PIP2‐NS3 interaction significantly reduces viral pathogenicity and provides protective efficacy in murine models, establishing PIP2‐mediated NS3 secretion as both a key virulence determinant and a promising ...

Development and Application of Attenuated Plant Viruses as Biological Control Agents in Japan

open access: yesViruses
In 1929, it was reported that yellowing symptoms caused by a tobacco mosaic virus (TMV) yellow mosaic isolate were suppressed in tobacco plants that were systemically infected with a TMV light green isolate.

A gorilla adenovirus-based vaccine against Zika virus induces durable immunity and confers protection in pregnancy [PDF]

open access: yes, 2019
The teratogenic potential of Zika virus (ZIKV) has made the development of an effective vaccine a global health priority. Here, we generate two gorilla adenovirus-based ZIKV vaccines that encode for pre-membrane (prM) and envelope (E) proteins (GAd-Zvp ...

Photodynamic Priming and Minocycline Overcome Chemoresistance by Reprogramming the Pancreatic Tumor Immune Microenvironment In Vivo

open access: yesAdvanced Science, EarlyView.
Dual priming with minocycline and photodynamic priming reprograms the pancreatic tumor microenvironment to overcome chemoresistance. By suppressing DNA repair enzyme Tdp1, inducing photooxidative damage, and enhancing irinotecan delivery via light‐activated nanoencapsulation, this strategy remodels the tumor microenvironment and drives immune ...

Synthetic O‐Polysaccharide Backbone Units for a Single Antigen Vaccine Against Two Major Non‐Typhoidal Salmonella Serovars

open access: yesAngewandte Chemie, EarlyView.
A modular strategy was developed to synthesize tri‐, hexa‐, and nona‐saccharides representing repeating units of the conserved O‐polysaccharide backbone of Salmonella. Conjugates of these glycans to bacteriophage Qβ induced strong IgG responses in mice and rabbits. Antibodies elicited recognized native O‐polysaccharides and conferred protection against

Research Progress on Varicella-Zoster Virus Vaccines

open access: yesVaccines
Varicella-zoster virus (VZV) poses significant public health challenges as the etiological agent of varicella (chickenpox) and herpes zoster (HZ), given its high transmissibility and potential for severe complications.

Rational Design of Broad‐Spectrum Anti‐Enteroviral Molecular Glues Targeting Enteroviral RNAi Suppressors

open access: yesAdvanced Science, EarlyView.
By leveraging this homodimerization mechanism, molecular glues were rationally designed to induce dysfunctional 3A dimerization, thereby restoring antiviral RNAi. The optimal molecular glue, VTP‐32, demonstrated potent and pan‐enterovirus (groups A, B, D) antiviral effects.

Recombinant Listeria promotes tumor rejection by CD8+ T cell-dependent remodeling of the tumor microenvironment. [PDF]

open access: yes, 2018
Agents that remodel the tumor microenvironment (TME), prime functional tumor-specific T cells, and block inhibitory signaling pathways are essential components of effective immunotherapy.
