The Inhibitory Effect of Validamycin A onAspergillus flavus [PDF]
Aspergillus flavusis one of the most common isolates from patients with fungal infections.Aspergillusinfection is usually treated with antifungal agents, but side effects of these agents are common. Trehalase is an essential enzyme involved in fungal metabolism, and the trehalase inhibitor, validamycin A, has been used to prevent fungal infections in ...

Response of microbial communities in the tobacco phyllosphere under the stress of validamycin. [PDF]
Validamycin, is classified as an environmentally friendly fungicide. It has high efficacy with little associated pollution risk, and it has been used in China on tobacco for many years especially during leaf spot season.

Chemical modification of validamycin A.
The chemical modification of validamycin A was examined. One of the synthetic validamycin analogs, N[(1S)-(1,4,6/5)-4,5,6-trihydroxy-3-hydroxymethyl-2-cyclohexenyl] [O-β-2-amino-2- deoxy-d-glucopyranosyl-(1-> 3)-(1S)-(1,2,4/3,5) -2,3,4- trihydroxy-5-hydroxymethyl-cyclohexy] amine monohydrochloride had almost the same activity against the sheath blight ...

Nucleotidylation of unsaturated carbasugar in validamycin biosynthesis [PDF]
Validamycin A is a member of microbial-derived C(7)N-aminocyclitol family of natural products that is widely used as crop protectant and the precursor of the antidiabetic drug voglibose. Its biosynthetic gene clusters have been identified in several Streptomyces hygroscopicus strains, and a number of genes within the clusters have been functionally ...

Catalytic Analysis of the Validamycin Glycosyltransferase (ValG) and Enzymatic Production of 4′′-epi-Validamycin A [PDF]
ValG is a glycosyltransferase (GT) that is responsible for the glucosylation of validoxylamine A to validamycin A. To explore the potential utilization of ValG as a tool for the production of validamycin analogues, a number of nucleotidyldiphosphate-sugars were evaluated as alternative substrates for ValG.

Biosynthetic studies on validamycins. I 1H and 13C NMR assignments of validamycin A.
The 1H and 13C NMR spectra of validamycin A have been assigned by use of 2D homonuclear correlation spectroscopy for protons, and off-resonance decoupling, single frequency off-resonance decoupling, and comparison studies with model compounds for carbons. Effects of pH on carbon chemical shifts have also been studied.

Microbial transformation of validamycins.
Alpha- or beta-Glucosidic linkage of validamycin was selectively cleaved by microbial hydrolysis and especially the conversion of validamycin C into validamycin A by the selective hydrolysis of alpha-glucoside linkage has important significance because validamycin C is considerably less active than validamycin A.

Effect of validamycins on glycohydrolases of Rhizoctonia solani.
The pseudo-oligosaccharides, validamycins, showed potent inhibitory activity against trehalase of Rhizoctonia solani while no significant inhibition was exhibited against cellulase, pectinase, chitinase, alpha-amylase, alpha- and beta-glucosidases. In particular, validoxylamine A strongly inhibited trehalase in a competitive manner with a Ki value of 1.
