IFI16 restricts SARS-CoV-2 replication by disrupting nucleocapsid-driven phase separation

open access: yesCommunications Biology
IFI16 is an interferon-inducible protein that senses viral DNA and can also restrict RNA virus replication. Here, using IFI16 knockout cellular models, we identify IFI16 as a host restriction factor that limits SARS-CoV-2 replication.

In Silico Analysis of Mutated SARS-CoV-2 N Protein of Java Origin and Human IgG Antibody as a Potential Vaccine Candidate [PDF]

open access: yesEPJ Web of Conferences
The high mutation rate of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) may alter viral protein structures and reduce the effectiveness of vaccines that primarily target the spike (S) protein.

Strategies for enhancing sensitivity in lateral flow assays

open access: yesVIEW, EarlyView.
Lateral flow assays (LFAs) are essential for cost‐effective, rapid point‐of‐care diagnostics. However, conventional colourimetric LFAs often lack the sensitivity required for low‐abundance biomarkers. This review evaluates six enhancement strategies: flow modulation, sample preconcentration, advanced reporters, chemical signal amplification, structural

Assembly of SARS-CoV-2 nucleocapsid protein with nucleic acid [PDF]

open access: yes
The viral genome of SARS-CoV-2 is packaged by the nucleocapsid (N-)protein into ribonucleoprotein particles (RNPs), 38 ± 10 of which are contained in each virion. Their architecture has remained unclear due to the pleomorphism of RNPs, the high

Naturally Occurring ZCCHC3 Variants Modulate Antiretroviral Activity in Cynomolgus Macaques

open access: yesMicrobiology and Immunology, EarlyView.
ABSTRACT Many mammalian cells restrict viral replication by utilizing various host restriction factors. We recently demonstrated that CCHC‐type zinc‐finger‐containing protein 3 (ZCCHC3) suppresses human immunodeficiency virus type 1 (HIV‐1) replication through multiple mechanisms.

Activation of PKR by Bunyamwera virus is independent of the viral interferon antagonist NSs [PDF]

open access: yes, 2003
Double-stranded RNA (dsRNA) is a by-product of viral RNA polymerase activity, and its recognition is one mechanism by which the innate immune system is activated.

Time‐Resolved Multi‐Omics Identify Biomarkers of Immediate Reactions to mRNA Vaccination

open access: yesAllergy, EarlyView.
We compared early vaccine‐induced immune and metabolic signatures and downstream immunity after second and booster BNT162b2 doses in participants with sARs, ISSRs, or no reactions. Neutrophil mobilization, antiviral programs, and robust anti‐S‐antibody responses occurred across reaction groups.

The Dynamic Landscape of Capsid Proteins and Viral RNA Interactions in Flavivirus Genome Packaging and Virus Assembly

open access: yesPathogens
The Flavivirus genus of the Flaviviridae family of enveloped single-stranded RNA viruses encompasses more than 70 members, many of which cause significant disease in humans and livestock.

Cell surface RNA virus nucleocapsid proteins: a viral strategy for immunosuppression?

open access: yesnpj Viruses
AbstractNucleocapsid protein (N), or nucleoprotein (NP) coats the genome of most RNA viruses, protecting and shielding RNA from cytosolic RNAases and innate immune sensors, and plays a key role in virion biogenesis and viral RNA transcription. Often one of the most highly expressed viral gene products, N induces strong antibody (Ab) and T cell ...
