Circulating tumor cell viability during and after radiotherapy mirrors treatment response in cancer patients

open access: yesMolecular Oncology, EarlyView.
Radiotherapy (RT) response depends on the DNA repair capacity of tumor and host cells. We show that circulating tumor cell (CTC) counts and apoptosis rates before and after RT predict treatment response and outcome, which can be accessed via easily accessible liquid biopsy approaches. Accelerated X-ray Structure Elucidation of a 36 kDa Muramidase/Transglycosylase Using wARP [PDF]

open access: yes, 1998
The X-ray structure of the 36kDa soluble lytic transglycosylase from Escherichia coli has been determined starting with the multiple isomorphous replacement method with inclusion of anomalous scattering at 2.7 Å resolution. Subsequently, before any model

Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Proteasomal degradation of intracellularly expressed Amblyomin‐X limits suicide gene therapy potential in melanoma cells

open access: yesFEBS Open Bio, EarlyView.
This study explores the feasibility of expressing the antitumoral protein Amblyomin‐X through a suicide gene therapy approach and investigates its intracellular fate after gene delivery. Although the gene is efficiently expressed, melanoma cells rapidly degrade the Amblyomin‐X protein via proteasome activity.

Single crystal x-ray diffraction studies on small, medium and large molecules [PDF]

open access: yes, 1996
Chapter 1. Production of crystals for diffraction analysis would be assisted by the devising of a set of rules which, given molecular formula, could predict crystal formation conditions.

EFEITO DO DIOXIDO DE CARBONO SOBRE A ESTABILIDADE DA FASE Ca2Fe2O5 APLICADA NO PROCESSO DE DEGRADAÇÃO FOTOCATALITICO DO AZUL DE METILENO

open access: yesQuímica Nova, 2015
Solid samples containing a Ca2Fe2O5 phase were synthesized using the Pechini method. The samples were characterized using X-ray diffraction, thermogravimetric analysis, differential thermal analysis, X-ray fluorescence, nitrogen adsorption/desorption ...

Evolutionarily divergent DUF4465 domains have a common vitamin B12‐binding function

open access: yesFEBS Open Bio, EarlyView.
We show that DUF4465 family proteins, widespread across bacteria from gut microbiomes, hydrothermal vents, and soil, share a common vitamin B12‐binding function. These augmented β‐jellyroll proteins bind vitamin B12 via extended loops. Our findings establish sequence‐diverse DUF4465 proteins as a widespread class of B12‐binding proteins, highlighting ...

A Phase-resolved View of Millihertz Quasiperiodic Oscillations in the Ultraluminous X-Ray Source M51 ULX-7: Evidence for a Magnetically Truncated Disk and Geometrical Beaming

open access: yesThe Astrophysical Journal
X-ray quasiperiodic oscillations (QPOs) are commonly observed in Galactic X-ray binaries and extragalactic ultraluminous X-ray sources (ULXs). In this study, we perform a phase-resolved analysis of recently discovered X-ray mHz QPOs in M51 ULX-7.

Optimizing photoactivation of PA‐mCherry for optical pooled CRISPR screens

open access: yesFEBS Open Bio, EarlyView.
Photoactivatable PA‐mCherry finds widespread use to optically tag individual cells. However, confocal 405 nm UV laser‐scanning (normal scan) is much less efficient than widefield UV illumination, limiting the use of PA‐mCherry on confocal instruments. We remedy this limitation by reporting that rapid and repeated confocal scanning with a low‐intensity,
