Analysis of Xanthophylls in Corn by HPLC

An HPLC method was developed using the C-30 carotenoid column to separate and identify the major xanthophylls in corn (lutein, zeaxanthin, and beta-cryptoxanthin). A photodiode array detector and a mobile phase consisting of methyl tert-butyl ether/methanol/water was used. All three xanthophylls eluted in less than 25 min.

Carotenol fatty acid esters in various fruits and vegetables are sources of carotenoids for humans and are cleaved during intestinal absorption to yield the parent xanthophyll. In the present study we demonstrate that low amounts of carotenol esters are present in human skin.

Orientation of xanthophylls in phosphatidylcholine multibilayers

Oriented multibilayers of dimyristoyl phosphatidylcholine (DMPC) modified with violaxanthin or zeaxanthin were examined by X-ray diffractometry and linear dichroism. It appears that pigment molecules and the normal to the bilayer plane form an angle of 24-25 degrees.

AbstractXanthophylls are dietary lipophilic compounds. Among them, lutein and zeaxanthin are the major carotenoids found in the human lens and retina, and referred as macular pigment within the retina. Lutein and zeaxanthin cannot be synthesized endogenously. They may therefore be considered as essential and must be provided by adequate dietary intakes.

IT is known1–3 that the chlorophylls (but not their pheophytins) will aggregate in a variety of non-polar solvents (for example, benzene and carbon tetrachloride), with characteristic spectra for the respective monomers and dimers. These aggregates are concentration-dependent in the usual mass action sense up to a concentration of at least 10−2 moles/l.
