Primary Megakaryocytes Reveal a Role for Transcription Factor Nf-E2 in Integrin αiibβ3 Signaling [PDF]

open access: yes, 1999
Platelet integrin αIIbβ3 responds to intracellular signals by binding fibrinogen and triggering cytoskeletal reorganization, but the mechanisms of αIIbβ3 signaling remain poorly understood.

From haemostasis to immunity: The expanding frontiers of platelet biology

open access: yesBritish Journal of Haematology, Volume 209, Issue 4, Page 1482-1485, October 2026.
Platelets are versatile cells whose functions extend far beyond haemostasis and thrombosis, actively regulating innate and adaptive immunity, angiogenesis, lymphangiogenesis, liver regeneration, atherosclerosis and tumour metastasis. This demonstrates their central role in various physiological processes and multiple life‐threatening diseases ...

A nonactivating ITGB3 mutation in the β3 cytoplasmic region causes macrothrombocytopenia with an impaired αIIbβ3/RhoA pathway

open access: yesBlood Vessels, Thrombosis & Hemostasis
: Almost all mutations of ITGA2B or ITGB3 identified in congenital macrothrombocytopenia induce constitutive activation of αIIbβ3. However, whether concomitant αIIbβ3 activation is essential for macrothrombocytopenia development remains unknown. Recently,

αIIbβ3 variants in ten families with autosomal dominant macrothrombocytopenia: Expanding the mutational and clinical spectrum.

open access: yesPLoS ONE, 2020
BackgroundRare pathogenic variants in either the ITGA2B or ITGB3 genes have been linked to autosomal dominant macrothrombocytopenia associated with abnormal platelet production and function, deserving the designation of Glanzmann Thrombasthenia-Like ...

Effects of the RGD loop and C-terminus of rhodostomin on regulating integrin αIIbβ3 recognition. [PDF]

open access: yesPLoS ONE, 2017
Rhodostomin (Rho) is a medium disintegrin containing a 48PRGDMP motif. We here showed that Rho proteins with P48A, M52W, and P53N mutations can selectively inhibit integrin αIIbβ3. To study the roles of the RGD loop and C-terminal region in disintegrins,

Harnessing blood clot as a native scaffold for orchestrating tissue repairs and regeneration

open access: yesBMEMat, Volume 4, Issue 3, September 2026.
The blood clot, owing to its dynamic composition and unique microenvironment, holds significant yet underappreciated potential for tissue engineering. This review systematically summarizes the pathophysiology of clot formation, the key regulatory factors shaping its microenvironment, and its applications in both pre‐clinical and clinical settings ...

Implications of the differing roles of the β1 and β3 transmembrane and cytoplasmic domains for integrin function

open access: yeseLife, 2016
Integrins are transmembrane receptors composed of α and β subunits. Although most integrins contain β1, canonical activation mechanisms are based on studies of the platelet integrin, αIIbβ3.

CD47 promotes mitogen‐activated protein kinase and epithelial‐to‐mesenchymal transition molecular programs to drive prometastatic phenotypes in non‐small cell lung cancer

open access: yesMolecular Oncology, Volume 20, Issue 8, Page 1963-1985, August 2026.
Beyond its role in immune evasion, this study identified that CD47 drives tumor‐intrinsic signaling in non‐small cell lung cancer (NSCLC). Transcriptomic profiling and functional studies revealed that CD47 regulates cell adhesion, migration, and metastasis through an ERK–EMT signaling axis.
