异基因造血干细胞移植(allo-HSCT)是目前高危组急性髓系白血病(AML)唯一可以根治的治疗手段[1]。然而,移植后复发仍是allo-HSCT后患者死亡的最主要原因[2]。通过微小残留病(MRD)监测及时发现MRD并给予抢先治疗可以有效预防移植后复发,提高长期生存率[3]。目前对于移植后MRD阳性患者的干预手段主要有供者淋巴细胞输注(DLI)、去甲基化治疗、靶向药物治疗、干扰素(IFN)等,但仍有部分患者疗效不佳[4-5]。

用Psi2STK/GCV对鼠C6神经胶质瘤细胞作用的初步研究

目的研究利用基因工程包装细胞进行基因转导和药物治疗相结合来治疗脑胶质瘤的可行性。方法利用分子生物学方法构建了逆转录病毒载体STK及基因工程包装细胞psi2STK,用psi2STK细胞和丙氧鸟苷(GCV)结合对胶质瘤细胞进行了基因治疗研究。结果C6胶质瘤细胞和基因工程包装细胞与不同浓度的丙氧鸟苷(GCV)一起培养,未感染的细胞比感染的细胞对丙氧鸟苷(GCV)更为敏感。结论通过体外实验证实:逆转录病毒转导HSV-TK基因在培养C6胶质瘤细胞中,提高了其对GCV的敏感性。
