壳聚糖纳米粒子基因载体的制备与相关研究

目的制备CS/pcDNA3.1/GDNF混悬液,并对壳聚糖作为基因载体的相关性能进行检测。方法采用复凝聚法制备CS/pcDNA3.1/GDNF混悬液,观察壳聚糖纳米粒子的物理性能,稳定性,基因结合效率,基因保护功能,缓慢释放效果及基因负载力和负载率。结果壳聚糖大小为106-228nm,性质稳定,具有良好的基因携带功能,能够有效地保护所携带基因,基因负载力及基因负载率较高。结论应用复凝聚法制得壳聚糖纳米粒子,能够满足基因载体的需要,是一个较理想的基因载体。
