D-缬氨酸抑制巨噬细胞M1极化的体外研究

目的研究D-缬氨酸对巨噬细胞M1极化的影响。方法在不同浓度的D-缬氨酸情况下,通过CCK-8方法测定分析细胞毒性,利用实时PCR分析多个基因的mRNA表达。然后通过Elisa方法检测IL-6,IL-1β,TNF-α的蛋白表达,使用荧光显微镜分析iNOS表达。结果 50mM D-缬氨酸对巨噬细胞没有细胞毒性,D-缬氨酸在mRNA和蛋白质水平下调IL-6,IL-1β和TNF-α的表达,细胞中iNOS也下调明显。结论 D-缬氨酸抑制RAW264.7巨噬细胞中LPS/IFN-γ诱导的M1巨噬细胞极化。
