Structure‐forward targeting of claudins with synthetic binders

open access: yesFEBS Letters, EarlyView.
Claudins form the paracellular barriers between epithelial and endothelial tissues at tight junctions and are targets for molecular binders with the goal of modulating barrier permeability. Claudin‐binding molecules are relevant in drug delivery or in altering claudin interactions with disease‐causing proteins.

PDK1 promotes breast cancer progression by enhancing the stability and transcriptional activity of HIF-1α

open access: yesGenes and Diseases
Pyruvate dehydrogenase kinase 1 (PDK1) phosphorylates the pyruvate dehydrogenase complex, which inhibits its activity. Inhibiting pyruvate dehydrogenase complex inhibits the tricarboxylic acid cycle and the reprogramming of tumor cell metabolism to ...

Nutrient/TOR signaling controls adipose mitochondrial transcription factor A (TFAM) to regulate organismal growth in Drosophila

open access: yesFEBS Letters, EarlyView.
Animals must match their growth rate to available nutrients. We show that in Drosophila larvae, the nutrient‐sensing TOR kinase controls growth by regulating levels of TFAM, a key regulator of mitochondrial function, in the adipose tissue. When nutrients are abundant, high TOR activity suppresses TFAM, lowering mitochondrial bioenergetic activity and ...

Hepatic Hypoxia-Inducible Factor 1α Mediates Ferroptosis via Transferrin Receptor 1 in Acute Liver Injury

open access: yesAntioxidants
Acute liver injury (ALI) is a potentially life-threatening condition lacking effective clinical drugs. Hypoxia-inducible factor-1α (HIF-1α) is a key regulator of both inflammation and metabolism.

Exploring the Glucocorticoid Actions of 1α-Hydroxycorticosterone (1α-OH-B) in the Elasmobranch Fishes [PDF]

open access: yes, 2014
The corticosteroid 1α-hydroxycorticosterone (1α-OH-B) is unique to the elasmobranch fishes. It is thought that 1α-OH-B acts as both the primary glucocorticoid (GC) and mineralocorticoid (MC) in these fishes, a dual role analogous to that of cortisol in ...

The effect of NT-PGC-1α on candidate gene expression in brown adipocytes.

open access: yes, 2016
(A) Schematic diagram of splicing events of the PGC-1α gene. Splicing between canonical exon 1a and exon 2 produces PGC-1α-a and NT-PGC-1α-a, whereas alternative splicing between distal exon 1b and exon 2 produces PGC-1α-b and NT-PGC-1α-b.

Studying the C1772T polymorphism of Hif-1α and TGF-β3 IVS5+104 A/G polymorphism in children with congenital non-syndromic neural tube defects and their mothers

open access: yesCellular and Molecular Biology, 2018
Prevalence of neural tube defect (NTD) has reduced after folic acid intake. However; which mechanisms are effective in NTD are not known exactly. In this study; due to the possible effects on hypoxic pathway and embryonic development, particularly on extracellular matrix components, Hif-1α Pro582Ser and TGF-β3 IVS5+104 A/G SfaN1 polymorphisms were ...

Autophagy and mitophagy in pancreatic β‐cell homeostasis and their involvement in diabetes pathophysiology

open access: yesFEBS Letters, EarlyView.
This review focuses on the role of autophagy and mitophagy in maintaining pancreatic β‐cell function and homeostasis. We discuss how genetic defects affecting these pathways contribute to the development of type 1, type 2, monogenic, and gestational diabetes. We further explore their potential as therapeutic targets. Created in BioRender.
