Chronobiology of Cancer: How Aging Fuels Oncogenesis at the Molecular Level
This graphical abstract illustrates the key biological pathways linking aging with cancer development and progression. In the upper left, cumulative exposure to ultraviolet radiation, toxins, and reactive oxygen species (ROS) causes DNA damage and genomic instability, whereas age‐related decline in repair mechanisms, such as ATM/ATR, BER, and NER ...

26S Proteasome: Structure and Function
The 26S proteasome is a molecular machine of approximately 2.5 MDa, which degrades ubiquitylated proteins in an ATP-dependent manner. It comprises two modules, the 20S proteasome and one or two copies of asymmetric 19S regulatory particles (RPs), which ...

Living donor heart valves offer superior durability for pediatric patients but trigger immune rejection. Lentiviral delivery of viral immune‐evasion genes US2 and Serpin b9 into donor valve cells and valve tissue suppresses HLA expression, reduces PBMC clustering and cytotoxic killing, and induces transcriptional immune‐tolerance programs, paving the ...

Role of S5b/PSMD5 in Proteasome Inhibition Caused by TNF-α/NFκB in Higher Eukaryotes
The ubiquitin-proteasome system is essential for maintaining protein homeostasis. However, proteasome dysregulation in chronic diseases is poorly understood.

Insights into the molecular architecture of the 26S proteasome [PDF]
Cryo-electron microscopy in conjunction with advanced image analysis was used to analyze the structure of the 26S proteasome and to elucidate its variable features. We have been able to outline the boundaries of the ATPase module in the “base” part of the regulatory complex that can vary in its position and orientation relative to the 20S core particle.

Difference between PA700-like proteasome activator complex and the regulatory complex dissociated from the 26S proteasome implies the involvement of modulating factors in the 26S proteasome assembly [PDF]
The PA700-like proteasome activator complex was highly purified from porcine erythrocytes, and its properties were compared with those of the regulatory complex disassembled from the purified 26S proteasome.

The role of TNFα in the process of renal allograft interstitial fibrosis is complex and multifaceted. As it can promote renal allograft interstitial fibrosis by inducing mitochondrial dysfunction and EndMT, and also mediating compensatory mitophagy through the NEDD4–HIF‐1α–BNIP3 pathway, which clears damaged mitochondria and inhibits EndMT, thereby ...
