A comprehensive method for extraction and quantitative analysis of sterols and secosteroids from human plasma

We describe the development of a method for the extraction and analysis of 62 sterols, oxysterols, and secosteroids from human plasma using a combination of HPLC-MS and GC-MS. Deuterated standards are added to 200 μl of human plasma.

The results showed that the reduction of plasma 24‐hydroxycholesterol (24‐OHC) was related to the cognitive decline in elderly surgical patients. Neuroinflammation induced by anesthesia and surgery significantly downregulated SREBP2 in hippocampal reactive astrocytes, resulting in reduced cholesterol synthesis.
