Introduction Autism Spectrum Disorder and Social Anxiety Disorder are mental illnesses characterized by a dysfunction in social behavior (SB); a phenomenon largely mediated by the medial prefrontal cortex (mPFC). Clinical studies have demonstrated that

Neuronal localization of the 5-HT2 receptor family in the amygdaloid complex
The amygdaloid complex (or amygdala), a heterogeneous structure located in the medial portion of the temporal lobe, is composed of deep, superficial, and remaining nuclei.

Role of 5-HT2A receptor antagonists in the treatment of insomnia
Insomnia encompasses a difficulty in falling asleep (sleep-onset insomnia) and/or a difficulty in staying asleep (SMI). Several selective serotonin-2A (5-HT2A) receptor antagonists have been in development as potential treatments for SMI. However, none have shown a sufficiently robust benefit-to-risk ratio, and none have reached market approval.

The T102C Polymorphism of the 5-HT2A-Receptor Gene in Fibromyalgia
Based on a possible involvement of serotonergic dysfunction in the pathophysiology of fibromyalgia (FM) and on preliminary reports of a possible genetically driven vulnerability for this disorder we investigated the silent T102C polymorphism of the 5-HT2A-receptor gene in 168 FM patients and 115 healthy controls.

Regulation of 5-HT2A receptor mRNA in P11 cells
P11 cells were used as an in vitro model system to explore effects of exposure to 5-HT on levels of mRNA encoding 5-HT2A receptors. Exposure of cells to the agonist 5-HT resulted in a doubling of receptor mRNA levels. mRNA levels returned to control levels within 8-16 h.

Enhanced effect of the hallucinogen DOI in L-DOPA receptor Gpr143-deficient mice
Hallucinogens, such as lysergic acid diethylamide, act primarily through the 5-hydroxytryptamine 2A (5-HT2A) receptor, but their regulatory mechanisms remain unclear.

Differential signaling signatures evoked by DOI versus lisuride stimulation of the 5-HT2A receptor
The 5-HT2A receptor is a target for hallucinogenic and non-hallucinogenic ligands that evoke unique behavioral, electrophysiological and molecular consequences.

The study employed a four‐tiered strategy: (1) UHPLC‐FTMS profiling of Citrus aurantium honey to characterize its chemical composition; (2) network pharmacology analysis integrating target prediction, protein–protein interaction networks, and KEGG pathway enrichment to identify the Thor1/Nprl2‐TORC1 axis as a key mechanistic pathway; (3) in vitro ...
