Loperamide Inhibits Tachykinin NK3-Receptor-Triggered Serotonin Release Without Affecting NK2-Receptor-Triggered Serotonin Release From Guinea Pig Colonic Mucosa

open access: yesJournal of Pharmacological Sciences, 2005
The effect of loperamide on tachykinin NK2- and NK3-receptor-mediated 5-HT outflow from guinea pig colonic mucosa was investigated in vitro. The selective tachykinin NK2-receptor agonist [β-Ala8]-neurokinin A4–10 (βAla-NKA) or the selective NK3-receptor ...

Structural insights into the ligand binding and Gi coupling of serotonin receptor 5-HT5A

open access: yesCell Discovery, 2022
5-hydroxytryptamine receptor 5A (5-HT5A) belongs to the 5-HT receptor family and signals through the Gi/o protein. It is involved in nervous system regulation and an attractive target for the treatment of psychosis, depression, schizophrenia, and ...

The 5-HT7 receptor as a potential target for treating drug and alcohol abuse [PDF]

open access: yes, 2015
Alcohol and drug abuse take a large toll on society and affected individuals. However, very few effective treatments are currently available to treat alcohol and drug addiction.

Engineering tandem VHHs to target different epitopes to enhance antibody‐dependent cell‐mediated cytotoxicity

open access: yesFEBS Open Bio, EarlyView.
Tandem VHH targeting distinct EGFR epitopes were engineered into a monovalent bispecific antibody (7D12‐EGA1‐Fc) with more potent ADCC without increasing affinity to EGFR. Structural modeling of 7D12‐EGA1‐Fc showed cross‐linking of separate EGFR domains to enhance CD16a engagement on NK cells.

Serotonin receptor‐mediated vasorelaxation occurs primarily through 5‐HT4 activation in bovine lateral saphenous vein

open access: yesPhysiological Reports
To better understand mechanisms of serotonin‐ (5‐HT) mediated vasorelaxation, isolated lateral saphenous veins from cattle were assessed for vasoactivity using myography in response to increasing concentrations of 5‐HT or selective 5‐HT receptor agonists.

Enhanced sensitivity of postsynaptic serotonin-1A receptors in rats and mice with high trait aggression [PDF]

open access: yes, 2001
Individual differences in aggressive behaviour have been linked to variability in central serotonergic activity, both in humans and animals. A previous experiment in mice, selectively bred for high or low levels of aggression, showed an up-regulation of ...

KLK7 overexpression promotes an aggressive phenotype and facilitates peritoneal dissemination in colorectal cancer cells

open access: yesFEBS Open Bio, EarlyView.
KLK7, a tissue kallikrein‐related peptidase, is elevated in advanced colorectal cancer and associated with shorter survival. High KLK7 levels in ascites correlate with peritoneal metastasis. In mice, KLK7 overexpression increases metastasis. In vitro, KLK7 enhances cancer cell proliferation, migration, adhesion, and spheroid formation, driving ...

Reactive oxygen species are required for 5-HT-induced transactivation of neuronal platelet-derived growth factor and TrkB receptors, but not for ERK1/2 activation. [PDF]

open access: yesPLoS ONE, 2013
High concentrations of reactive oxygen species (ROS) induce cellular damage, however at lower concentrations ROS act as intracellular second messengers. In this study, we demonstrate that serotonin (5-HT) transactivates the platelet-derived growth factor

Zebrafish studies identify serotonin receptors mediating antiepileptic activity in Dravet syndrome. [PDF]

open access: yes, 2019
Dravet syndrome is a life-threatening early-onset epilepsy not well controlled by antiepileptic drugs. Drugs that modulate serotonin (5-HT) signalling, including clemizole, locaserin, trazodone and fenfluramine, have recently emerged as potential ...

Discriminating depression, physical and social anhedonia by neurotransmitter related challenge tests [PDF]

open access: yes, 2016
The aim of this study was to investigate, if anhedonia, a salient component of depression, shows similar response patterns to neurotransmitter challenge tests as depression, and if the two questionnaire based components Physical (PA) and Social (SA ...
