Radiosensitization by Quaternary Salts of 5-nitroimidazole Derivatives

The radiosensitizing effects of five newly synthesized quaternary salts of 5-nitroimidazole derivatives on the survival of TC-SV40 mammalian cells have been measured. A toxicity study was carried out in order to determine the concentrations to be used in the radiosensitizing experiments.

Cross-resistance of trichomonads to 5-nitroimidazole-derivatives

The strainT. foetus KV 1/M 100, a sub-strain ofT. foetus Kv 1, was made highly resistant to metronidazole by several passages in mice under increasing drug pressure. This drug resistance was accompained by a remarkable cross-resistance to all 5-nitroimidazole-derivatives tested in vitro and in vivo.

Genetics of Resistance of Bacteroides Species to 5-Nitroimidazole

Thirty-three clinical isolates of Bacteroides strains with a moderate-to-high level of resistance to 5-nitroimidazole were compared with three strains previously studied by Southern blotting of their DNA. Whether plasmid-borne or chromosomally determined, all genetic determinants that are resistant to 5-nitroimidazole belong to two hybridization groups.

Studies on the disposition of a 5-nitroimidazole in laboratory animals

The disposition and metabolism of a 5-nitroimidazole compound (SC 28538) was investigated in the rat, beagle dog and rhesus monkey. The absorption of [14C]-SC 28538 was rapid and essentially complete after oral dosage in male animals, and also after intravaginal dosage in the female rat. Peak plasma levels of radioactivity occurred within 2 h of dosage.

Moxnidazole [3-(5-nitrol-1-methyl-2-imidazolyl)-methyleneamino-5-morpholinomethyl-2-oxazolidinone, HCl], known to be mutagenic in microbial tests and Drosophila, induced genetic alterations in somatic cells of mice. Ethyl methanesulfonate (EMS) served as positive control.

Effects of niridazole and 5-nitroimidazoles on heart mitochondrial respiration

2.3 micromoles/mg protein of MFNI induced a 60% decrease in the heart mitochondrial ADP-stimulated oxygen uptake using glutamate-malate as substrate. The same amount of niridazole, ipronidazole, DA 3851 and ornidazole led to falls of less than 20% in the oxygen uptake, whilst metronidazole was ineffective.
