The Role of the N-Domain in the ATPase Activity of the Mammalian AAA ATPase p97/VCP [PDF]
p97/valosin-containing protein (VCP) is a type II ATPase associated with various cellular activities that forms a homohexamer with each protomer containing an N-terminal domain (N-domain); two ATPase domains, D1 and D2; and a disordered C-terminal region.

AAA+ ATPase chaperone p97/VCPFAF2 governs basal pexophagy
Peroxisomes are organelles that are central to lipid metabolism and chemical detoxification. Despite advances in our understanding of peroxisome biogenesis, the mechanisms maintaining peroxisomal membrane proteins remain to be fully elucidated.

SPG4 Hereditary Spastic Paraplegia: From Etiology to Therapy
Abstract Hereditary spastic paraplegias (HSPs) comprise a heterogeneous group of heritable neurodegenerative disorders resulting from mutations in a wide variety of genes. HSP locomotor symptoms include lower limb weakness and spasticity that arise from progressive degeneration of corticospinal axons projecting from the motor cortex to the distal ...

AAA+ proteins carry out diverse functions in cells. In most cases, their ATPase activity is tightly regulated by protein partners and target ligands, but the mechanism for this control has remained unclear. We have identified a conserved link between the

Role of AAA-ATPase Cdc48p in peroxisomal quality control [PDF]
Most peroxisomal matrix proteins contain a type 1 peroxisomal targeting signal (PTS1), which is recognized by the cytosolic receptor Pex5p for delivery into peroxisomes. Following cargo translocation, the receptor is monoubiquitinated and recycled to the cytosol by the AAA-ATPases Pex1p and Pex6p.

A Perspective on the Enhancer Dependent Bacterial RNA Polymerase
Here we review recent findings and offer a perspective on how the major variant RNA polymerase of bacteria, which contains the sigma54 factor, functions for regulated gene expression.

Resolving the TorsinA Oligomerization Conundrum: The Glycan Hypothesis
TorsinA is a AAA+ ATPase involved in the severe neurological disease Early Onset Torsion Dystonia. Despite the impressive progress in the field in the recent years, the structural organization and function of this intriguing molecule is still not clear ...

Genomic Signatures of Endemic Pathogen Exposure in a Declining Amphibian
ABSTRACT Endemic wildlife diseases can impose strong selection on threatened populations, yet whether persistence is associated with adaptive genomic responses remains poorly understood. We tested whether long‐term exposure to Batrachochytrium dendrobatidis (Bd) following population declines has left detectable selection signatures in the genome of the
