Development of visible light‐sensitive human neuropsin (OPN5) via single amino acid substitution

open access: yesFEBS Letters, EarlyView.
The present study determines a key amino acid residue, Lys91, for defining UV sensitivity of human OPN5. Heterologous action spectroscopy of the wild type and K91 mutants of OPN5 in HEK293T cells reveals that substitution of Lys91 with neutral (alanine) or acidic amino acids (glutamic or aspartic acids) causes substantial shifts in spectral sensitivity

Disruption of type 3 adenylyl cyclase expression in the hypothalamus leads to obesity [PDF]

open access: yes, 2016
Evidence from human studies and transgenic mice lacking the type 3 adenylyl cyclase (AC3) indicates that AC3 plays a role in the regulation of body weight. It is unknown in which brain region AC3 exerts such an effect.

Targeting AAV vectors

open access: yesMolecular Therapy, 2003
One of the many hurdles that face gene therapists is the requirement to optimize gene delivery systems in such a way that both efficient and selective transgene production is achieved in vivo within a chosen cell or tissue. Exploiting native vector tropism can sometimes achieve this goal irrespective of additional modifications. For example, adenoviral

Syngeneic AAV Pseudo-particles Potentiate Gene Transduction of AAV Vectors

open access: yesMolecular Therapy - Methods & Clinical Development, 2017
Adeno-associated virus (AAV) vectors have emerged as a safe and efficient gene therapy platform. One complication is that a significant amount of empty particles have always been generated as impurities during AAV vector production. However, the effects of such particles on AAV vector performance remain unclear.

Discovery and Treatment of Action Potential‐Independent Myotonia in Hyperkalemic Periodic Paralysis

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective Hyperkalemic periodic paralysis (hyperKPP) is characterized by attacks of transient weakness. A subset of hyperKPP patients suffers from transient involuntary contraction of muscle (myotonia). The goal of this study was to determine mechanisms causing myotonia in hyperKPP.

Intracisternal delivery of NFkappaB-inducible scAAV2/9 reveals locoregional neuroinflammation induced by systemic kainic acid treatment.

open access: yesFrontiers in Molecular Neuroscience, 2014
We have previously demonstrated disease-dependent gene delivery in the brain using an AAV vector responding to NFB activation as a probe for inflammatory responses.

The Potential of eCD4-Ig, Delivered by Adeno-Associated Virus (AAV) Vector as a Novel Vaccine for HIV/AIDS Infection

open access: yesScripta Score Scientific Medical Journal, 2021
Background: HIV/AIDS has already become one of the world's major health issues taking its toll on millions of lives each year. Developing an HIV vaccine with excellent efficacy has become a global urgency that must be addressed immediately.

Lack of complex I activity in human cells carrying a mutation in MtDNA-encoded ND4 subunit is corrected by the Saccharomyces cerevisiae NADH-quinone oxidoreductase (NDI1) gene [PDF]

open access: yes, 2001
The gene for the single subunit, rotenone-insensitive, and flavone-sensitive internal NADH-quinone oxidoreductase of Saccharomyces cerevisiae (NDI1) can completely restore the NADH dehydrogenase activity in mutant human cells that lack the essential ...

Comprehensive AAV capsid fitness landscape reveals a viral gene and enables machine-guided design

open access: yesScience, 2019
The fitness landscape of AAV Adeno-associated virus (AAV) is an important gene therapy vector. Using tools from synthetic biology, Ogden et al. provide a comprehensive view of how sequence changes in capsid proteins affect AAV properties.

A Vertically‐Stacked Optoelectronic Sensor for Localized Hemodynamics Monitoring

open access: yesAdvanced Functional Materials, EarlyView.
This work introduces a hemodynamics monitoring sensor that features vertically stacked microLEDs and a heterogeneously integrated photodetector. The vertically stacked microLEDs enable localized measurements, and by designing the interoptode distance according to the depth of the target region, this vertically stacked optoelectronic sensor is ...
