LIBX‐A401: A Novel Selective Inhibitor of Acyl‐CoA Synthetase Long Chain Family Member 4 (ACSL4) and Its Binding Mode

The discovery of LIBX‐A401, a potent and selective ACSL4 inhibitor derived from rosiglitazone is reported. HDX‐MS reveals that LIBX‐A401 stabilizes the ACSL4 C‐terminus and alters the fatty acid (FA) gate domain. Consistently, photoaffinity labeling, molecular dynamics, and mutagenesis identify key binding residues within the FA pocket.

The structure and biosynthetic origin of the enigmatic “yellow affinity substance” (YAS) from the anaerobe thermophile Clostridium thermocellum (now: Acetivibrio thermocellus) were elucidated. YAS is a complex of unusual antibacterial arylpolyene alkaloids (celluxanthenes) that are produced by various cellulolytic anaerobes.

A Unique THN Motif Is Critical for Enabling Efficient C‐Terminal Traceless Cleavage

THN is identified as the key motif to facilitate the rapid traceless cleavage of gp41‐1. The deflection of THN motif not only avoids the interaction between C‐exteins and active sites, but also separates Asp107 away from Asn125. In addition, a flexible block F contributes to faster C‐cleavage.
