Genotyping the ADAMTS5-J (Deltagen) mouse strain v1 [PDF]
ADAMTS5, the main aggrecanase, is a protein that seems to play a functional role in the development of brown and white adipose tissue (WAT) and browning of WAT. These observations were made using the ADAMTS5-P mice originally generated by Pfizer, in collaboration with Lexicon Genetics (The Woodlands, TX, USA).

The extracellular matrix (ECM) protease Adamts5 and its ECM substrates are critical regulators of inflammation and fibrosis; whether Adamts5 also regulates muscle regeneration is not known. Right tibialis anterior (TA) muscles from adult Adamts5--/- mice

ABSTRACT Background Adolescent Idiopathic Scoliosis (AIS) is a spinal deformity of unknown etiology that begins in the intervertebral disc and emerges during pubertal growth. In the vertebral growth plate, adjacent to the disc, powerful growth plate morphogens (GPMs) modulate this growth. We evaluated whether GPMs can also alter the capacity of annulus

miR-21-5p protects IL-1β-induced human chondrocytes from degradation
Objective Osteoarthritis (OA) is a prevalent degenerative disease caused by various factors. MicroRNAs are important regulators in OA. MiR-21-5p expression is decreased in OA cartilage, but the effects of modulating miR-21-5p on cartilage regeneration ...

ABSTRACT Equine melanocytic neoplasms (EMN) are aggressive tumours characterised by high metastatic potential and limited therapeutic options available. However, the molecular mechanisms underlying their progression remain poorly understood. This study therefore presents the integrative phosphoproteomic analysis of EMN tissue, with the aim of ...

An adhesive hydrogel depot with polyphenol‐armored nanogenes is developed for treating aged osteoarthritis. This system maintains miR‐140 bioactivity, prolongs its intra‐articular retention, and enhances transfection into chondrocytes, leading to the downregulation of senescence‐associated pathways.

Anti-ADAMTS5 monoclonal antibodies: implications for aggrecanase inhibition in osteoarthritis [PDF]
The extracellular matrix of articular cartilage is structurally specialized for efficient absorption of mechanical impact. In particular, giant aggregates of the large chondroitin sulfate proteoglycan, aggrecan, with the glycosaminoglycan, hyaluronan, allow cartilage to resist compressive load.

Lactational lactoferrin deficiency exerts lasting effects on epididymal adipose tissue development from lactation into adulthood: it impairs adipocyte hyperplasia and induces pathological hypertrophy, resulting in lower body weight yet exacerbated metabolic dysfunction under a high‐fat diet in adulthood.

• ADAMTS4 is induced primarily in cardiac fibroblasts during sepsis. • ADAMTS4 cleaves TSP1 at the 236–246 aa functional region. • TSP1 cleavage activates TGF‐β/Smad and NF‐κB signalling in fibroblasts. • Targeting ADAMTS4 reduces fibroblast activation and cardiac fibrosis.
