PRMT5/Sohlh2/Sirt1 Signaling Pathway in Vascular Endothelial Cells Modulates Lung Metastasis of Triple‐Negative Breast Cancer

open access: yesAdvanced Science, EarlyView.
The cover image shows that vascular endothelial cells play an important role in lung metastasis of TNBC. Endothelial Sohlh2 acts as a gatekeeper against TNBC lung metastasis by limiting endothelial activation and tumor cell passage. PRMT5 reduces Sohlh2 stability, weakening this protective barrier.

Molecular Analysis of N6-Methyladenine Patterns in \u3cem\u3eTetrahymena thermophila\u3c/em\u3e Nuclear DNA [PDF]

open access: yes, 1989
We have cloned two DNA fragments containing 5\u27-GATC-3\u27 sites at which the adenine is methylated in the macronucleus of the ciliate Tetrahymena thermophila. Using these cloned fragments as molecular probes, we analyzed the maintenance of methylation

Palmitic Acid Promotes Antiviral Innate Immunity via ZDHHC20‐Mediated CMPK2 Palmitoylation

open access: yesAdvanced Science, EarlyView.
Metabolites have important functions in innate immune activation and regulation. Wang et al. uncover metabolic regulation of antiviral immunity through CMPK2 palmitoylation, which regulates CMPK2 mitochondrial localization and is promoted by ZDHHC20 but reversed by PPT1, inhibition of which antagonizes viral infection in mice.

Mechanisms of the action of adenine on anti‐allergic effects in mast cells

open access: yesImmunity, Inflammation and Disease, 2018
Introduction Mast cells play an important role in allergic responses. Methods We herein demonstrated the mechanisms of inhibitory effect of adenine on IgE/antigen‐induced degranulation and TNF‐α release in mast cells.

Site-Specific Methylation of Adenine in the Nuclear Genome of a Eucaryote, \u3cem\u3eTetrahymena thermophila\u3c/em\u3e [PDF]

open access: yes, 1986
DNA in the polyploid macronucleus of the ciliated protozoan Tetrahymena thermophila contains the modified base N6-methyladenine. We identified two GATC sites which are methylated in most or all of the 45 copies of the macronuclear genome.

Mechanozyme: An Artificial Enzyme With a Mechanophore Framework

open access: yesAdvanced Science, EarlyView.
Mechanical destabilization of a G‐quadruplex mechanophore in a DNAzyme markedly boosts its catalytic activity, demonstrating “mechanozymes” as artificial enzymes whose functions are modulated by mechanical stress exerted by ultrasonication. This mechanozyme principle is broadly applicable to control biomolecular activities.

WTAP‐Mediated m6A Modification Targets the LRP1‐Lipid Metabolism Axis to Regulate Joint Cartilage Regeneration

open access: yesAdvanced Science, EarlyView.
WTAP drives cartilage regeneration by activating an LRP1‐dependent lipid metabolic program in macrophages, enhancing IL‐10 and TGF‐β secretion to promote chondrogenic differentiation. Leveraging this mechanism, virtual screening identifies LRP1‐targeting compounds that effectively stimulate cartilage repair, highlighting a druggable epigenetic ...

Influence of a fluorobenzene nucleobase analogue on the conformational flexibility of RNA studied by molecular dynamics simulations [PDF]

open access: yes, 2006
Chemically modified bases are frequently used to stabilize nucleic acids, to study the driving forces for nucleic acid structure formation and to tune DNA and RNA hybridization conditions.

Rewiring NADH Metabolism Through NQO1‐Mediated Redox Cycling for Targeted Follicular Lymphoma Therapy

open access: yesAdvanced Science, EarlyView.
ABSTRACT Follicular lymphoma (FL) remains an incurable B‐cell malignancy with high relapse rates, and conventional therapies are often limited by significant toxicity, highlighting the need for novel treatments. We identified that FL exhibits markedly low expression of NAD(P)H: quinone oxidoreductase 1 (NQO1), a key enzyme required for activating ...

Metabolism‐Based Biomarkers for Rapid Phenotypic Antibiotic Susceptibility Testing

open access: yesAdvanced Science, EarlyView.
Metabolic differences emerging after antibiotic exposure provide rapid phenotypic signals of susceptibility and resistance. This review summarizes metabolic biomarkers for rapid phenotypic AST, associated with nutrient uptake, respiratory activity, metabolic reprogramming, and enzymatic function. ABSTRACT The accelerating global crisis of antimicrobial
