NAADP+ synthesis from cADPRP and nicotinic acid by ADP-ribosyl cyclases

Biochemical and Biophysical Research Communications, 2006
ADP-ribosyl cyclases (ADPRCs) are present from lower Metazoa to mammals and synthesize the Ca2+-active (di)nucleotides cyclic ADP-ribose (cADPR), NAADP+, and ADP-ribose (ADPR), involved in the regulation of important cellular functions. NAADP+ can be synthesized by ADPRCs from NADP+ through a base-exchange reaction, which substitutes nicotinamide for ...

Unifying mechanism for Aplysia ADP-ribosyl cyclase and CD38/NAD+ glycohydrolases

Biochemical Journal, 2000
Highly purified Aplysia californica ADP-ribosyl cyclase was found to be a multifunctional enzyme. In addition to the known transformation of NAD+ into cADP-ribose this enzyme is able to catalyse the solvolysis (hydrolysis and methanolysis) of cADP-ribose.

Endogenous ADP-ribosylation of Gs subunit and autonomous regulation of adenylate cyclase

Nature, 1986
Although cholera toxin induces a marked stimulation of adenylate cyclase activity in rat adipocyte plasma membranes, the holotoxin induces only a slight increase of cyclic AMP accumulation in intact cells. A similar apparent anomaly is seen with pertussis toxin, which has been shown to inhibit the Gi subunit of adenylate cyclase, and has a greater ...

Altered Regulation of Adenylate Cyclase after Toxin-Catalyzed ADP-Ribosylation

1985
Publisher Summary This chapter presents an analysis of three types of ADP-Ribosylation, namely, ADP-ribosylation of Gs by choleragen, ADP-ribosylation of Gi by pertussis toxin, and ADP-ribosylation of transducin. The other known ADP-ribosyltransferases are bacteriophage products or bacterial toxins whose functions should probably be described as ...

Cyclic ADP-ribose, the ADP-ribosyl cyclase pathway and calcium signalling

Molecular and Cellular Endocrinology, 1994
Cyclic adenosine diphosphate-ribose, an endogenous metabolite of nicotinamide adenine dinucleotide was first characterized as a potent Ca2+ mobilizing agent in sea urchin eggs. Mounting evidence points to it being an endogenous activator of Ca(2+)-induced Ca2+ release by non-skeletal muscle ryanodine receptors in several invertebrate and mammalian cell
