Elucidation of degradation pattern and immobilization of a novel alginate lyase for preparation of alginate oligosaccharides

International Journal of Biological Macromolecules, 2020
Alginate lyases are important enzymes to prepare alginate oligosaccharides for industrial applications and elucidating the degradation pattern of alginate lyases is essential for expanding their applications. Herein, we cloned and expressed AlyPL6, a novel member of polysaccharide lyase family-6 (PL6) with high activity from Pedobacter hainanensis NJ ...

Effect of γ-Irradiation on Degradation of Alginate

open access: yesJournal of Agricultural and Food Chemistry, 2003
The aqueous solution of alginate was irradiated by 60Co gamma-rays in the dose range of 10-500 kGy. To assess the effect of irradiation on the degradation of alginate, the irradiation-induced changes in the viscosity, molecular weight, color, monomer composition, and sequence were measured.

Radiation-induced degradation of sodium alginate

Polymer Degradation and Stability, 2000
Abstract Alginates were irradiated as solids or in aqueous solution with Co 60 gamma rays in the dose range of 20 to 500 kGy to investigate the effect of radiation on alginates. Degradation was observed both in the solid state and solution. The degradation in solution was remarkably greater than that in the solid.

Modification of alginate degradation properties using orthosilicic acid

open access: yesJournal of the Mechanical Behavior of Biomedical Materials, 2012
Biopolymers such as alginates have been widely researched for clinical use. Their clinical application, however, have been limited due to their unpredictable and often rapid degradation rates. Here we show that the degradation of an alginate hydrogel can be tailored through the addition of orthosilicic acid (OSA).

Encapsulation of fibroblasts causes accelerated alginate hydrogel degradation

open access: yesActa Biomaterialia, 2010
Calcium-alginate hydrogel has been widely studied as a material for cell encapsulation for tissue engineering. At present, the effect that cells have on the degradation of alginate hydrogel is largely unknown. We have shown that fibroblasts encapsulated at a density of 7.5 x 10(5) cells ml(-1) in both 2% and 5% w/v alginate remain viable for at least ...

Gut bacterial alginate degrading enzymes

Essays in Biochemistry, 2023
AbstractAlginates are abundant marine anionic polysaccharides consumed by humans. Thus, over the years some understanding has emerged about alginate utilization by human gut microbiota (HGM). However, insights have been obtained only recently at the molecular level with regard to structure and function of alginate degrading and metabolizing enzymes ...

Characterization of Multiple Alginate Lyases in a Highly Efficient Alginate-Degrading Vibrio Strain and Its Degradation Strategy

Applied and Environmental Microbiology, 2022
Alginate is an important carbon source in the marine environment, and vibrios are major alginate utilizers. Previous studies focused only on the characteristics of individual alginate lyases in vibrios, but few of them discussed the comprehensive alginate-degrading strategy.

The depolymerization of sodium alginate by oxidative degradation

Pharmaceutical Development and Technology, 2011
Alginate has been extensively used as a carrier for macromolecules and as gene delivery vehicle. Both properties are molecular weight (Mw) dependent. Herein, we investigated factors affecting the oxidative depolymerization of alginate. The depolymerization process occurred mainly in the first 1 h.

Alginate Sequencing: An Analysis of Block Distribution in Alginates Using Specific Alginate Degrading Enzymes

Biomacromolecules, 2011
Distribution and proportion of β-D-mannuronic and α-L-guluronic acid in alginates are important for understanding the chemical-physical properties of the polymer. The present state of art methods, which is based on NMR, provides a statistical description of alginates.
