GCN2 in proteostasis: structural logic, signalling networks and disease

Threats to protein synthesis activate the kinase GCN2, initiating the integrated stress response (ISR). GCN2 is triggered by stalled ribosomes and uncharged tRNAs, which accumulate when amino acids are scarce. The ISR adjusts cellular physiology by promoting redox balance, protein quality control, and mitochondrial optimisation.

Quantitative analysis of DNA‐GATA1 binding alterations linked to hematopoietic disorders

Native holdup allows the quantitative determination of affinities between full‐length transcription factors and DNA. Mutations in either the protein or the DNA can modulate binding strength, which can be precisely quantified using this approach. Applied to GATA1, it revealed mutations that alter DNA binding.

Functional and transcriptomic analyses in Neurospora crassa reveal a crucial role for the N‐glycoprotein deglycosylation process in fungal homeostasis

In Neurospora crassa, a cytosolic GH18 endo‐β‐n‐acetylglucosaminidase (ENGase) functions as an active deglycosylating enzyme. Deletion of this severely alters tolerance to multiple stress conditions and causes extensive transcriptomic reprogramming, including upregulation of a GH20 exo‐β‐n‐acetylhexosaminidase under ER stress, indicating that N. crassa

Proteomic Analysis of Serum Extracellular Vesicles Revealed Fibulin‐3 as a Novel Predictive Biomarker for Hepatocellular Carcinoma in Patients With Chronic Hepatitis B

We carried out proteomic analysis of serum extracellular vesicles to identify biomarkers predictive of hepatocellular carcinoma (HCC) development in patients with chronic hepatitis B receiving nucleos(t)ide analog therapy. Candidate proteins identified through proteomic analysis were subsequently quantified in serum by ELISA.

A posttranslational proteomic survey of a single anatomically preserved human 20‐week postconception brain

A well‐preserved 20 post‐conception week human brain was obtained and finely dissected into 18 anatomically distinct regions, including the pia mater. Each region underwent in‐depth proteomic analysis, encompassing both total protein content and post‐translational modifications.
