A radioimmunoassay for determination of 3 alpha-hydroxy-5 alpha-pregnan-20-one (allopregnanolone) in serum or plasma has been developed and evaluated. The method employs rabbit antiserum to 3 alpha-hydroxy-5 alpha-pregnane-11,20-dione-11-O-carboxymethyloxime bovine serum-albumin conjugate and tritiated radioligand.

7-Nor-20-oxopregn-5-en-3β-yl acetate was converted into (20R)-5β,6β-epoxy-7-nor-5β-pregnane-3β,20-diyl diacetate in three steps. Stereospecific migration of the 6α-hydride ion led to a 6-oxo derivative with a 5α-configuration. The (Z)-oxime of this ketone underwent Beckmann rearrangement to yield a lactam with the nitrogen in position 7.

Flumazenil blocks the anxiolytic action of allopregnanolone

In the burying behaviour test allopregnanolone (0.5 mg/rat) reduced the cumulative time of burying, interpreted as a reduction in anxiety. The selective benzodiazepine antagonist, flumazenil (5 and 10 mg/kg), did not affect the burying behaviour when administered alone but effectively prevented the reduction produced by allopregnanolone.
