The role of allopregnanolone in depression and anxiety
Progress in Neurobiology, 2014Neuroactive steroids such as allopregnanolone do not only act as transcriptional factors in the regulation of gene expression after intracellular back-oxidation into the 5-α pregnane steroids but may also alter neuronal excitability through interactions with specific neurotransmitter receptors.

Effects of PhD examination stress on allopregnanolone and cortisol plasma levels and peripheral benzodiazepine receptor density [PDF]
Contains fulltext : 58623.pdf (Publisher’s version ) (Open Access)Peripheral benzodiazepine receptor (PBR) density in blood platelets and plasma allopregnanolone concentration in humans were determined following acute stress as ...

Allopregnanolone in Women with Premenstrual Syndrome
Hormone and Metabolic Research, 19985Alpha-reduced metabolites of progesterone, especially 3alpha-hydroxy-5alpha-pregnan-20-one (allopregnanolone) are now listed to neurosteroids. Their anesthetic properties can be explained by their allosteric interaction with GABA(A) receptors. A woman's organism is provided with an abundance of progesterone during the luteal phase of the menstrual ...

A novel radioimmunoassay of allopregnanolone
Steroids, 1995A radioimmunoassay for determination of 3 alpha-hydroxy-5 alpha-pregnan-20-one (allopregnanolone) in serum or plasma has been developed and evaluated. The method employs rabbit antiserum to 3 alpha-hydroxy-5 alpha-pregnane-11,20-dione-11-O-carboxymethyloxime bovine serum-albumin conjugate and tritiated radioligand.

Regenerative potential of allopregnanolone
Brain Research Reviews, 2008The neuroendocrine status of the brain has been linked to the quality of the aging process, to the risk of Alzheimer's disease and to progression of neurodegenerative pathology. Data from multiple levels of analysis ranging from in vitro cellular models to in vivo animal models to clinical investigations indicate that the decline of neurosteroids play ...

Neurosteroids: 7-aza-allopregnanolone—a poor substitute for allopregnanolone
Tetrahedron, 20077-Nor-20-oxopregn-5-en-3β-yl acetate was converted into (20R)-5β,6β-epoxy-7-nor-5β-pregnane-3β,20-diyl diacetate in three steps. Stereospecific migration of the 6α-hydride ion led to a 6-oxo derivative with a 5α-configuration. The (Z)-oxime of this ketone underwent Beckmann rearrangement to yield a lactam with the nitrogen in position 7.

Alcohol, allopregnanolone and aggression in mice
Psychopharmacology, 2000Aggressive behavior of certain individual animals can be greatly increased when under the influence of low doses of alcohol. One of alcohol's neurochemical actions that may be relevant to alcohol-heightened aggression (AHA) is its positive modulation of the GABA(A) receptor complex. The objective of this study was to investigate whether alcohol ...

Background: Neurosteroids modulate epileptic activity by interacting with the γ-aminobutyric acid type A receptor, but their brain levels are still undetermined. Objectives: We aimed to establish neurosteroid levels in the neocortex and hippocampus by liquid chromatography/mass

Flumazenil blocks the anxiolytic action of allopregnanolone
European Journal of Pharmacology, 1995In the burying behaviour test allopregnanolone (0.5 mg/rat) reduced the cumulative time of burying, interpreted as a reduction in anxiety. The selective benzodiazepine antagonist, flumazenil (5 and 10 mg/kg), did not affect the burying behaviour when administered alone but effectively prevented the reduction produced by allopregnanolone.
