A zebrafish model carrying an identical human RHO S334X allele reveals two independent genetic layers shaping retinitis pigmentosa (RP) severity: a protective 3‐bp cis‐regulatory insertion that attenuates transgene expression, and a dominant trans‐acting modifier that restores a severe phenotype.

HRM analysis using different amplicon lengths.
Multiple amplicon lengths (90, 129, and 212 bp) were tested for mutation analyses in order to optimize the HRM analysis. First row: -d1 curve; second row: -d2 curve. The 90-bp amplicon showed the most interpretable heteroduplex-derived peaks.

ABSTRACT Flavivirus subgenomic RNAs (sfRNAs) antagonise antiviral defences, yet how sfRNAs are organized and maintained in cells remains poorly understood. Here we identify ubiquitin C‐terminal hydrolase L3 (UCHL3) as a post‐translational regulator of flavivirus sfRNA stability and function.
