The liver‐brain axis: A multidimensional regulatory network implicated in Alzheimer's disease pathogenesis and clinical implications

open access: yesAnimal Models and Experimental Medicine, EarlyView.
Schematic diagram of the core pathways of the liver‐brain axis in regulating AD. The liver regulates cerebral Aβ deposition, tau phosphorylation, and neuroinflammation through pathways such as metabolic detoxification (urea cycle, ketone body metabolism, glutathione antioxidant system), molecular secretion (APOE, CRP, FGF21, IGF‐1), and Aβ clearance ...

Establishing Sensory Neurons as Therapeutic Targets in Peripheral Neuropathy Driven by Polyglutamine Expanded Murine ATXN3

open access: yesAnnals of Neurology, EarlyView.
Repeat expansion disorders frequently involve peripheral neuropathy, yet mechanisms remain unclear. Using a spinocerebellar ataxia type 3 (SCA3) Knock‐In Atxn3Q300/Q6, we identify progressive sensorimotor deficits, peripheral nerve pathology, and dorsal root ganglia RNA splicing dysregulation.

Advancements in Alzheimer's Disease Therapy: The Role of Anti-Amyloid Monoclonal Antibodies - A Literature Review

open access: yesJournal of Education, Health and Sport
Alzheimer's disease is a neurodegenerative disorder characterized by progressive cognitive decline and neuronal damage. Anti-amyloid monoclonal antibodies such as aducanumab, lecanemab, and donanemab, represent the first disease-modifying therapies for ...

White matter hyperintensities in Alzheimer's disease in the era of anti‐amyloid therapies

open access: yesAlzheimer’s & Dementia: Diagnosis, Assessment & Disease Monitoring
White matter hyperintensities (WMHs) are highly prevalent in Alzheimer's disease (AD) and arise from interacting vascular pathologies (including hypertensive small vessel disease and cerebral amyloid angiopathy) alongside inflammatory and ...

Multimodal Characterization of Glymphatic‐Related Magnetic Resonance Imaging Markers in Huntington's Disease: A Multi‐Cohort Retrospective Study

open access: yesAnnals of Neurology, EarlyView.
Objective To characterize magnetic resonance imaging (MRI)‐based glymphatic surrogates in Huntington's disease (HD) using MRI measures of perivascular diffusivity and structural perivascular alterations across multiple large cohorts. Methods We analyzed 2,731 MRI sessions from 880 participants across 3 large retrospective HD cohorts.

Indole‐3‐Acetic Acid Protects Bovine Hepatocytes Against Palmitic Acid‐Induced Lipid Accumulation and ER Stress Through SIRT3

open access: yesAnimal Research and One Health, EarlyView.
Cows with ketosis during the perinatal period often experience damage to the liver, a core metabolic organ, thereby exacerbating the disease's inherent vicious cycle. This study indicates that serum 3‐IAA concentrations were reduced in ketotic cows and show a significant negative correlation with hepatic TG content.

The Centiloid Scale in Amyloid PET Imaging: Current Role in Alzheimer’s Disease Diagnosis, Treatment Planning, and Monitoring During Anti-Amyloid Therapy: A Clinical Perspective

open access: yesDiagnostics
Amyloid positron emission tomography (PET) has become a critical tool in the diagnosis and treatment of Alzheimer’s disease (AD). The Centiloid (CL) scale, a tracer/scanner-independent, standardized quantification unit introduced in 2015, transforms ...

Extracellular Vesicles‐Based Biological Macromolecule Delivery Systems: Biogenesis, Characterization, and Emerging Clinical Applications

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Extracellular vesicles (EVs) are nanoscale membrane‐bound structures that play a pivotal role in intercellular communication by transporting bioactive molecules, including proteins, lipids, nucleic acids, and organelles, between cells. Originating from the endosomal pathway, EVs reflect the physiological and pathological status of their parent

How many sites are enough? a novel, site-based power analysis method for real-world registry studies of anti-amyloid monoclonal antibodies

open access: yesJAR Life
Background: Real-world registries ALZ-NET (US) and AD-DMT (Japan) support safety surveillance of anti-amyloid antibodies. Conventional power calculations—dividing required patients by mean per-site caseload—can underestimate the number of centers needed ...
