Amyloid beta-protein precursor-rich platelet microparticles in thrombotic disease.
Thrombosis and haemostasis, 1995We investigated the association of amyloid beta-protein precursor (APP) and platelet derived microparticles in 20 normal controls and 91 patients with various diseases causing a thrombotic tendency. Compared with the controls, the mean percentage of APP-positive microparticles was significantly greater in the patients with cerebral infarction (39.1 +/-

Thiophilic Interaction Chromatography (TIC) of Amyloid-β Protein Precursor
Journal of Alzheimer's Disease, 2007Neuritic plaques, one of the diagnostic characteristics of an AD, contain extracellular deposits of amyloid-β (Aβ) derived from amyloid-β protein precursor (AβPP). The objective of this study was to extract AβPP out of HEK293 cells and to purify it. Two procedures were chosen for purification of AβPP: Thiophilic Interaction Chromatography (TIC) and ...

Magnesium Modulates Amyloid-β Protein Precursor Trafficking and Processing
Journal of Alzheimer's Disease, 2010Alzheimer's disease (AD), the most common form of dementia, is characterized by the presence of excessive deposits of aggregated amyloid-β (Aβ), which is derived from the amyloid-β protein precursor (AβPP) following processing by β- and γ-secretase. Metal elements are implicated in the pathophysiology of AD.

Cytoplasmic Tail Adaptors of Alzheimer's Amyloid-β Protein Precursor
Molecular Neurobiology, 2004Alzheimer's disease is characterized pathologically by senile plaques in the brain. The major component of senile plaques is amyloid-beta (Abeta), which is cleaved from Alzheimer's Abeta protein precursor (AbetaPP). Recently, information regarding the cytoplasmic tail of AbetaPP has started to emerge, opening up various insights into the physiological ...

Amyloid β-protein precursor is associated with extracellular matrix
Brain Research, 1990The ultrastructural distribution of the amyloid beta-protein precursor (ABPP) in cultures of a neuronal cell line is examined. ABPP is associated with the extensive extracellular matrix (ECM) secreted by these cells. The possible role of matrix associated ABPP in the pathogenesis of Alzheimer's disease is discussed.

Caspase cleavage of the amyloid precursor protein modulates amyloid β‐protein toxicity
Journal of Neurochemistry, 2003AbstractThe amyloid β‐protein precursor (APP) is proteolytically cleaved to generate the amyloid β‐protein (Aβ), the principal constituent of senile plaques found in Alzheimer's disease (AD). In addition, Aβ in its oligomeric and fibrillar forms have been hypothesized to induce neuronal toxicity.

To observe the expression of amyloid beta precursor protein and amyloid beta protein in neurons of hippocampal CA1 region after brain ischemia in human.Neuronal damage was examined by using HE staining, and expression of APP and Abeta(1-40) was determined by immunohistochemistry in the hippocampal CA1 region of the brain specimens of 43 patients who ...

Pathogenic Effects of Cerebral Amyloid Angiopathy Mutations in the Amyloid β‐Protein Precursor
Annals of the New York Academy of Sciences, 2002Abstract: Cerebral amyloid β‐protein angiopathy (CAA) is a key pathological feature of patients with Alzheimer's disease and certain related disorders. Several mutations have been identified within the Aβ region of the Aβ protein precursor (AβPP) gene that appear to enhance the severity of CAA.

Complementary DNA for the mouse homolog of the human amyloid beta protein precursor
Biochemical and Biophysical Research Communications, 1987The human amyloid beta protein is a major component of brain amyloid found in patients with Alzheimer's disease. As an initial step to understand the biological function of its precursor protein, we have isolated cDNA for the mouse homolog of the human beta protein precursor. Comparison of the predicted amino acid sequence with that of human revealed a

Proteolytic processing of amyloid β protein precursor (APP) by thrombin
Biochemical and Biophysical Research Communications, 1992Search for proteases responsible for an altered processing of APP which generates intermediates containing beta/A4 peptide is preceding to understand the formation of beta amyloid deposits characteristic of Alzheimer's disease, since many studies reveal that APP is ordinarily processed so as not to generate beta amyloid.
