Nonsteroidal Anti-Inflammatory Drugs and Ectodomain Shedding of the Amyloid Precursor Protein [PDF]

open access: yes, 2009
Background: Epidemiological studies have suggested that long-term use of nonsteroidal anti-inflammatory drugs (NSAIDs) is associated with a reduced incidence of Alzheimer's disease (AD).

Exercise for the prevention of Alzheimer's disease: Multiple pathways to promote non-amyloidogenic AβPP processing

open access: yesAging and Health Research, 2022
With advancements in our ability to treat or manage more diseases than ever, people are able to live longer in greater health. However, for individuals who develop Alzheimer's disease (AD), therapeutic options are currently limited to symptom management ...

Regulation of Amyloid β-Protein Precursor by Phosphorylation and Protein Interactions [PDF]

open access: yesJournal of Biological Chemistry, 2008
Amyloid beta-protein precursor (APP), a type I membrane protein, is cleaved by primary alpha-or beta-secretase and secondary gamma-secretase. Cleavage of APP by beta- and gamma-secretases generates amyloid beta-protein, the main constituent of the cerebrovascular amyloid that accompanies Alzheimer disease. The generation and aggregation of amyloid beta-

In‐Depth Cell‐Type‐Specific Proteome Landscape of the Brain from Human Amyloid‐β Overexpression Mouse Model

open access: yesAdvanced Science, EarlyView.
In situ Cell‐type‐specific proteome Analysis using antibody‐mediated Biotinylation (iCAB) is a method that directly targets and analyzes proteins in specific cell types within tissue. Applied to 5xFAD mice, iCAB identifies ≈8000 cell‐type‐specific proteins, revealing unique pathways in neurons, astrocytes, and microglia.

Circadian modulation of microglial physiological processes and immune responses

open access: yesGlia, Volume 71, Issue 2, Page 155-167, February 2023., 2023
Main Points Microglia are rhythmic cells with an oscillatory expression in clock genes, cytokines, and other microglial markers. Microglial circadian rhythms determine their physiological function and immune responses. Abstract Microglia is considered the central nervous system (CNS) resident macrophages that establish an innate immune response against

LRP1 Has a Predominant Role in Production over Clearance of Aβ in a Mouse Model of Alzheimer's Disease. [PDF]

open access: yes, 2019
The low-density lipoprotein receptor-related protein-1 (LRP1) has a dual role in the metabolism of the amyloid precursor protein (APP). In cellular models, LRP1 enhances amyloid-β (Aβ) generation via APP internalization and thus its amyloidogenic ...

Neuronal C/EBPβ Shortens the Lifespan via Inactivating NAMPT

open access: yesAdvanced Science, EarlyView.
Schematic diagram illustrating the mechanism of shortened lifespan in Thy 1‐C/EBPβ transgenic mice and intervention strategies. Briefly, C/EBPβ/AEP escalation in the aging brain triggers NAMPT fragmentation and NAD+ depletion. Knockout of AEP from Thy 1‐C/EBPβ Tg/Tg mice ameliorates cognitive impairments and elongates the lifespan.

Deletion of MyD88 in astrocytes prevents β‐amyloid‐induced neuropathology in mice

open access: yesGlia, Volume 71, Issue 2, Page 431-449, February 2023., 2023
Main Points MyD88 is upregulated in astrocytes in Alzheimer's disease. Deletion of MyD88 in astrocytes protects against β‐amyloid‐induced synaptic toxicity and cognitive impairment in mice. Abstract As the understanding of immune responses in Alzheimer's disease (AD) is in its early phases, there remains an urgency to identify the cellular and ...
