Purification of Rat Angiotensinogen

A method of purifying rat angiotensinogen in three chromatography steps with a yield 3-4 times better than previous methods is described. Using chromatofocusing media for two steps and DEAE-affigel blue for the third step it was possible to separate angiotensinogen into three major peaks with pI of 5.25 (peak B), 4.80 (peak C) and 4.50 (peak D).

Abstract A four step procedure has been devised for the isolation of human angiotensinogen. Antisera to human angiotensinogen has been prepared. It appears to be an immunochemically homogenous protein. Some heterogeneity can be noted on polycrylamide electrophoresis. Clearance of rabbit plasma angiotensinogen and relationship to CSF angiotensinogen

Rabbit plasma angiotensinogen was purified 1,390-fold by classical purification procedures. Analytical polyacrylamide gel electrophoresis and direct activity assay revealed that the purified preparations were greater than 90% native angiotensinogen.
