Cell-Cycle-Associated Amplified Genomic-DNA Fragments (CAGFs) Might Be Involved in Chloroquine Action and Resistance in Plasmodium falciparum [PDF]

open access: yesarXiv, 2017
As a cheap and safe antimalarial agent, chloroquine (CQ) has been used in the battle against malaria for more than half century. However, the mechanism of CQ action and resistance in Plasmodium falciparum remains elusive. Based on further analysis of our published experimental results, we propose that the mechanism of CQ action and resistance might be ...

Hybrid Molecules as Efficient Drugs against Multidrug‐Resistant Malaria Parasites

open access: yesChemMedChem, EarlyView.
Among hybrid molecules currently developed as antimalarial drug candidates, emoquine‐1 exhibits high activity against all the multidrug‐resistant Plasmodium strains tested up to now, including artemisinin‐resistant quiescent parasites, critical parameters for promising antimalarial drugs. It is also curative in mouse malaria.

Novel quinazolinones active against multidrug‐resistant Mycobacterium tuberculosis: synthesis, antimicrobial evaluation and in silico exploration of PonA1 as a potential target

open access: yesChemMedChem, Accepted Article.
Quinazolinone derivatives have emerged as promising scaffolds in antimicrobial drug discovery. This work focuses on the design, synthesis, and evaluation of novel quinazolinone‐based compounds and predicts their potential to interact with mycobacterial penicillin‐binding proteins (PBPs).

Nucleus may be the key site of chloroquine antimalarial action and resistance development [PDF]

open access: yesarXiv, 2017
The first proposed hypothesis about the mechanism of chloroquine (CQ) action on malaria parasites is DNA intercalation hypothesis which indicates that the site of CQ action is within the nucleus. Later on the interest of research was shifted from nucleus to lysosome due to the report of CQ accumulation within lysosome.

Comparative Computational Study on the Robinson–Gabriel Synthesis and Bischler–Napieralski Reaction: Density Functional Theory Investigation of T3P‐Mediated Ring Closure

open access: yesEuropean Journal of Organic Chemistry, EarlyView.
The detailed comparison of the Bischler–Napieralski and Robinson–Gabriel reactions of tryptamides using propylphosphonic anhydride (T3P) that leads to different products in case of a small change in the substrate is provided. The computed mechanistic pathways explain why the oxoalkyl tryptamide derivative selectively reacts in a Robinson–Gabriel ...
