Engineering of Peptide‐Inserted Base Editors with Enhanced Accuracy and Security
This work developed a novel peptide insertion strategy to enhance the performance of base editors. Through changing the composition and position of the inserted peptide, peptide‐inserted base editors are engineered to exhibit robust on‐target editing activity and improved precision.

APOBEC3 in papillomavirus restriction, evolution and cancer progression
The APOBEC3 family of cytidine deaminases plays critical roles in host innate immunity. These enzymes function as viral DNA mutators capable of restricting an array of virus infections [1]. Given the potent antiviral function of APOBEC3s, strategies to enhance their activity have been suggested as a novel class of antiviral agents.

Characterization of Vif domains that mediate Feline Immunodeficiency Virus antagonism of APOBEC3-H and APOBEC3-CH restriction [PDF]
AbstractFeline immunodeficiency virus (FIV) Vif mediates degradation of two anti-lentiviral feline APOBEC3 (fA3) proteins, fA3Z3 and fA3Z2bZ3. HIV-1 Vif targets the restriction factor human APOBEC3G (A3G, hA3Z2g-Z1c) for proteasome degradation to mediate viral evasion. Despite this similarity, FIV and HIV-1 Vif share limited homology.

The Intricate Interplay between APOBEC3 Proteins and DNA Tumour Viruses
APOBEC3 proteins are cytidine deaminases that play a crucial role in the innate immune response against viruses, including DNA viruses. Their main mechanism for restricting viral replication is the deamination of cytosine to uracil in viral DNA during ...

Monkeypox Virus: WHO's Second Public Health Emergency of International Concern Within 2 Years
The figure shows examples of zoonotic diseases and their affected populations. The red arrows indicate the path of monkeypox virus infections first from a wild animal reservoir (rodents, squirrels) as a zoonosis mostly to children in central and west Africa, followed by the 2022/3 epidemic with human‐to‐human transmission in men having sex with men and

Evolvability of cancer-associated genes under APOBEC3A/B selection
Summary: Evolvability is an emergent hallmark of cancer that depends on intra-tumor heterogeneity and genetic variation. Mutations generated by APOBEC3 contribute to genetic variation and tumor evolvability.

Spatial immune heterogeneity in a mouse tumor model after immunotherapy
Spatial immune heterogeneity in the tumor microenvironment and its drivers in a mouse model of tumors induced by human papillomaviruses were investigated following immunotherapy. Gene expression was determined by RNA sequencing and mutations by whole exome sequencing in different tumor areas.

Temporal proteomic analysis of HIV infection reveals remodelling of the host phosphoproteome by lentiviral Vif variants [PDF]
Viruses manipulate host factors to enhance their replication and evade cellular restriction. We used multiplex tandem mass tag (TMT)-based whole cell proteomics to perform a comprehensive time course analysis of >6500 viral and cellular proteins during ...

The Role of the Urea Cycle in the Alzheimer's Disease Brain
Alzheimer's Disease (AD) is the leading cause of dementia, characterized by the accumulation of β‐amyloid plaques and neurofibrillary tangles in the brain. Despite extensive research, these hallmarks have not yet yielded effective therapies. Recent studies associate metabolic dysfunctions with AD, including elevated urea and ammonia, suggestive of a ...

APOBEC3 Hypermutations in HIV-1 Infected Cases in Turkey
Host genetic factors may play an effective role on the human immunodeficiency virus (HIV) pathogenesis. APOBEC3 (apolipoprotein B mRNA editing enzyme catalytic polypeptide like-3) proteins are cellular antiviral proteins which inhibits HIV replication in the absence of vif (virion infectivity factor).
