Improved Aptamers for the Diagnosis and Potential Treatment of HER2-Positive Cancer

open access: yesPharmaceuticals, 2016
Aptamers provide a potential source of alternative targeting molecules for existing antibody diagnostics and therapeutics. In this work, we selected novel DNA aptamers targeting the HER2 receptor by an adherent whole-cell SELEX approach.

Allosteric DNAzyme‐Enabled Sensitive and Multiplex Detection of Biomarkers for Rapid Diagnosis of Urinary Tract Infections

open access: yesAdvanced Science, EarlyView.
A catalytic allosteric DNAzyme assay (SMART) is developed by engineering DNAzyme into a unimolecular biosensor, enabling highly sensitive and multiplex detection of small molecules and nucleic acids. SMART enables extraction‐free, one‐pot, one‐step, isothermal, and cost‐effective detection of biological markers, achieving >95% accuracy for rapid ...

Aptamers for CD Antigens: From Cell Profiling to Activity Modulation

open access: yesMolecular Therapy: Nucleic Acids, 2017
Nucleic acid-based aptamers are considered to be a promising alternative to antibodies because of their strong and specific binding to diverse targets, fast and inexpensive chemical synthesis, and easy labeling with a fluorescent dye or therapeutic agent.

The development of aptamer-based probes for the detection of TB antigens ESAT-6.CFP-10 potential TB diagnostic tools

open access: yes, 2013
Includes abstract.Includes bibliographical references.Lack of point-of-care (PoC) diagnostic tools for TB hinders control of the disease, particularly in resource-limited, high HIV and TB prevalence countries. Therefore, there is a need for simple, rapid,

Aptamer‐Directed Porous DNA Nanocomposite Hydrogel for Active Pulp Preservation: Immunomodulation, Stem Cell Recruitment and Reparative Dentinogenesis

open access: yesAdvanced Science, EarlyView.
This study presents an injectable DNA‐based porous hydrogel integrating catechol motifs and targeting aptamers for pulpitis management. Upon in situ crosslinking, the scaffold actively recruits endogenous dental pulp stem cells, restores redox homeostasis, and modulates immune responses.

Bilateral SERS‐Microneedle Patch for Co‐Diagnosis of Diabetes Mellitus and Tuberculosis Comorbidity

open access: yesAdvanced Science, EarlyView.
Our study introduced a U@mP@Au‐mediated bilateral SERS‐microneedle patch for analyzing glucose levels from ISF and sensitively detecting TB biomarkers from sputum, respectively. This integrated dual‐analyte detecting platform can be used for co‐diagnosis of diabetes mellitus (DM) and tuberculosis (TB) comorbidity, which was demonstrated in ex vivo ...

Aptamer‐Targeted PrPC Drives Colorectal Cancer Metastasis via a LYN‐STAT3 Complex and Enables Liquid Biopsy Detection

open access: yesAdvanced Science, EarlyView.
The aptamer WHY‐3E identifies PrPC as a CRC driver. Stabilized by USP18, endocytosed PrPC forms a LYN/STAT3 complex, upregulating MSN transcription to promote metastasis. Crucially, WHY‐3E sensitively detects PrPC‐positive circulating exosomes, establishing a robust theoretical foundation for non‐invasive clinical diagnostics.
