Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

Sea ice friction and Arctic sea ice dynamics

open access: yes, 2010
A series of double-shear experiments were undertaken in the Environmental Test Basin at the Hamburgische Shiffbau-Versuchsanstalt (HSVA) in Hamburg. The aim of these experiments was to improve understanding of transient behaviour in sea ice friction. To this end we have fitted our experimental results to a rate- and state- dependent model of friction ...

MITF maintains genome stability in nonmelanocyte lineages

open access: yesMolecular Oncology, EarlyView.
MITF is essential for melanocyte survival and acts as an oncogene in 10%–20% of melanomas. We show that MITF depletion causes genome instability in nonmelanocytic cells, leading to LATS2‐mediated P53 activation, cell cycle arrest, and apoptosis. This study highlights the role of MITF as a genome maintenance factor beyond the melanocyte lineage. Created

On the interannual variability of arctic sea‐level pressure and sea ice

open access: yes, 1992
Monthly mean sea‐level pressure (SLP) data from the Northern Hemisphere for the period January 1952‐December 1987 are analysed. Fluctuations in this field over the Arctic on interannual time‐scales and their statistical association with fluctuations ...

A novel quinazolinone insulin receptor inhibitor and its synergy with an EGFR inhibitor in glucose‐driven glioblastoma

open access: yesMolecular Oncology, EarlyView.
The novel styrylquinazolinone‐based molecule W1B effectively suppresses glioblastoma by inhibiting IGF1R and EGFR. In high‐glucose microenvironments driving tumor resistance, W1B acts synergistically with the EGFR inhibitor dacomitinib. This combination safely blocks compensatory survival signaling in zebrafish xenograft models. Showcasing promising in

Oncogenic DMTF1β promotes cancer cell motility by regulating autophagy through ULK1 stabilization

open access: yesMolecular Oncology, EarlyView.
In the current study, we demonstrate that the oncogene DMTF1β regulates ULK1 stability by reducing its proteasomal degradation in cancer cells. This stabilization enables ULK1 to induce autophagy, which in turn facilitates cancer cell migration. Consequently, reduced DMTF1β levels lead to decreased autophagy and impaired cancer cell migration.

The Arctic Winter Sea Ice Quadrupole Revisited

open access: yes, 2017
International audienceThe dominant mode of Arctic sea ice variability in winter is often maintained to be represented by a quadrupole structure, comprising poles of one sign in the Okhotsk, Greenland and Barents Seas, and opposing sign in the Labrador ...

Patient therapy outcome modeling in cancer organoids is improved by cancer‐associated fibroblasts and organoid assembly convolution

open access: yesMolecular Oncology, EarlyView.
Patient‐derived organoids (PDOs) from pancreatic, colorectal, and gastric cancers were used to evaluate standard and experimental therapies. Incorporating cancer‐associated fibroblasts (CAFs) into organoid cultures improved patient therapy outcome prediction.

Analysis of CryoSat’s radar altimeter waveforms over different Arctic sea ice regimes [PDF]

open access: yes, 2014
Satellite altimetry has been used to derive information about sea ice thickness in the Arctic already for several decades. As part of the algorithms applied the shape of the radar signal is used to identify leads, the open water between ice floes ...
