Arginine deprivation and autophagic cell death in cancer [PDF]

open access: yesProceedings of the National Academy of Sciences, 2014
Our increasing awareness over the last decade, that reprogramming of cellular energetics is a key hallmark of tumorigenesis, has reinvigorated the search for novel tractable metabolic targets in cancer cells (1). From innovative diagnostics based on the tracer 18F-deoxyglucose in positron emission tomography (FDG-PET) to the development of next ...

The Effect of Arginine Deprivation on the Replication of Vaccinia Virus

open access: yesJournal of General Virology, 1971
Summary Arginine has been shown to be essential for the replication of vaccinia virus in HeLa cell cultures. The yield of infective virus was dependent upon the concentration of arginine in the medium, maximum yield being obtained with 0.09 mm-arginine.

Intracellular arginine-dependent translation sensor reveals the dynamics of arginine starvation response and resistance in ASS1-negative cells

open access: yesCancer & Metabolism, 2021
Background Many cancers silence the metabolic enzyme argininosuccinate synthetase 1 (ASS1), the rate-limiting enzyme for arginine biosynthesis within the urea cycle. Consequently, ASS1-negative cells are susceptible to depletion of extracellular arginine

Cytotoxicity of tumor antigen specific human T cells is unimpaired by arginine depletion. [PDF]

open access: yes, 2013
Tumor-growth is often associated with the expansion of myeloid derived suppressor cells that lead to local or systemic arginine depletion via the enzyme arginase.

Cytotoxicity of Arginine Deprivation to AML Cells is Mediated by Autophagy

open access: yes, 2020
In this study, we assess the activation of autophagy in AML cells following arginine deprivation, the mechanism of its activation, and its impact on cell cytotoxicity.

Anti-tumor activity of arginine deiminase via arginine deprivation in retinoblastoma

open access: yesOncology Reports, 2007
In spite of recent advances in the treatment of retinoblastoma, chemotherapy is still challenging in high-stage intraocular retinoblastoma or metastatic retinoblastoma. Here, we investigated whether arginine deprivation via arginine deiminase (ADI) could be a new anti-tumor therapy in retinoblastoma cells.

Effects of the combined arginase and canavanine treatment on leukemic cells in vitro and in vivo [PDF]

open access: yesThe Ukrainian Biochemical Journal, 2016
It was previously demonstrated in in vitro experiments that canavanine (Cav), a natural toxic arginine analogue of plant origin, is a promising candidate for augmenting the antineoplastic effects of arginine starvation.

Arginine deprivation response is mediated by MPK2.

open access: yes, 2016
Promastigotes of L. mexicana mutants lacking both copies of the MPK2 gene (Δmpk2) and those containing a single wild type copy of the gene re-inserted into their genome (addback) were subjected to 2 hours arginine deprivation as described in Fig 1.

Vascular Effects of Arginine Vasopressin during Fluid Deprivation in the Rat [PDF]

open access: yesJournal of Clinical Investigation, 1981
The vascular effects of arginine vasopressin (AVP) were examined in conscious Sprague-Dawley rats. In six control rats, synthetic AVP at a dose of 40 ng/kg, injected as an intravenous bolus, resulted in a rise in mean arterial blood pressure (BP) from 127 to 149 mm Hg (P < 0.005).

Next generation oncolytic viruses expressing PADI1 and TIMP2 exhibit anti-tumor activity against melanoma in nude and humanized mouse models

open access: yesMolecular Therapy: Oncolytics, 2023
Immunotherapy of metastatic melanoma (MM) has vastly improved the longevity of only a minority of patients. To broaden the repertoire of agents against MM, we investigated the effectiveness of locally interrupting tumor blood endothelial cell ...
