Pancreatic cancer cell lines deficient in argininosuccinate synthetase are sensitive to arginine deprivation by arginine deiminase [PDF]
Abstract Eukaryotic cells can synthesize the non‐essential amino acid arginine from aspartate and citrulline using the enzyme argininosuccinate synthetase (ASS). It has been observed that ASS is underexpressed in various types of cancers ASS, for which arginine become auxotrophic.

Dissecting the Adaptive Response to Arginine Deprivation in Hodgkin Lymphoma
Abstract Background In Hodgkin Lymphoma (HL), neoplastic cells orchestrate an inflammatory microenvironment leading to sterile inflammation, T-cell anergy, and immune deficiency. Our group showed that in HL patients the aminoacid degrading enzyme Arginase-1 is increased, associated with poor outcome, and leads to arginine (Arg ...

Amino acid deprivation induces AKT activation by inducing GCN2/ATF4/REDD1 axis
Amino acid availability is sensed by various signaling molecules, including general control nonderepressible 2 (GCN2) and mechanistic target of rapamycin complex 1 (mTORC1).

LMF2 drives progression and capecitabine resistance in triple‐negative breast cancer (TNBC). Mechanistically, VCP prevents NBR1‐mediated autophagic degradation of LMF2, thereby stabilizing it. Stabilized LMF2 subsequently suppresses NDP52‐mediated autophagic degradation of HMGCS1, a rate‐limiting cholesterol biosynthesis enzyme.

Rapidly growing tumor cells require a nutrient-rich environment in order to thrive, therefore, restricting access to certain key amino acids, such as arginine, often results in the death of malignant cells, which frequently display defective cell cycle ...

VCIPIP1 emerges as a molecular link between protein stability and prostate cancer progression. By deubiquitinating and stabilizing UBE3A, VCIPIP1 promotes p53 degradation and progression to castration‐resistant prostate cancer. Elevated VCIPIP1 expression is associated with shorter castration‐resistant prostate cancer–free survival, highlighting the ...

When nucleus pulposus cells senesce, PHGDH decreases, reducing serine synthesis and glutathione, impairing antioxidant capacity. The intermediate α‐KG also drops, lowering JMJD3 demethylase activity, increasing H3K27me3 at the Rap1b promoter, and reducing Rap1b expression, whose down‐regulation in turn activates mTORC1 and suppresses autophagy ...

CKMT1 inhibits VDAC1 oligomerization via a dual‐region mechanism: its NTD interacts with the VDAC1 C‐terminal domain, while residues 1–39 sterically hinder dimerization, thereby reducing mtDNA leakage and AIM2 inflammasome activation, suppressing neuronal pyroptosis and protecting against cerebral ischemic injury. ABSTRACT Ischemic stroke is one of the

Objective To identify autoantibodies in presymptomatic individuals that associate with the onset of rheumatoid arthritis (RA) and to distinguish early RA from osteoarthritis (OA), particularly in individuals lacking classic RA serologic markers. Methods We analyzed serum and plasma from three cohorts: presymptomatic individuals who later developed RA ...
