ATG8 localization in apicomplexan parasites [PDF]

open access: yesAutophagy, 2014
The ATG genes are highly conserved in eukaryotes including yeasts, plants, and mammals. However, these genes appear to be only partially present in most protists. Recent studies demonstrated that, in the apicomplexan parasites Plasmodium (malaria parasites) and Toxoplasma, ATG8 localizes to the apicoplast, a unique nonphotosynthetic plastid with 4 ...

CHARACTERIZATION OF ATG8 IN LEPIDOPTERAN INSECT CELLS

Archives of Insect Biochemistry and Physiology, 2013
Yeast Atg8 and mammalian microtubule‐associated protein light chain 3 (LC3) are landmark proteins essential for autophagy. Here the lepidopteran Atg8, a homolog of LC3, is characterized. Sequence analysis reveals that Atg8 proteins are highly conserved in lepidopteran species.

For the last time, it is GFP-Atg8, not Atg8-GFP (and the same goes for LC3) [PDF]

open access: yesAutophagy, 2011
GFP-Atg8/LC3 is a very useful marker to follow macroautophagy (hereafter autophagy). In contrast, Atg8/LC3-GFP has relatively limited uses. Unfortunately, many authors do not seem to appreciate the difference between these two constructs. I hope to put an end to that now.

Mechanisms and Pathophysiological Roles of the ATG8 Conjugation Machinery [PDF]

open access: yesCells, 2019
Since their initial discovery around two decades ago, the yeast autophagy-related (Atg)8 protein and its mammalian homologues of the light chain 3 (LC3) and γ-aminobutyric acid receptor associated proteins (GABARAP) families have been key for the tremendous expansion of our knowledge about autophagy, a process in which cytoplasmic material become ...

Non-canonical roles of ATG8 for TFEB activation

Biochemical Society Transactions, 2022
Autophagy is an evolutionally conserved cytoplasmic degradation pathway in which the double membrane structure, autophagosome sequesters cytoplasmic material and delivers them to lysosomes for degradation. Many autophagy related (ATG) proteins participate in the regulation of the several steps of autophagic process.

In Vitro Reconstitution of Atg8 Conjugation and Deconjugation

2017
Macroautophagy, hereafter autophagy, is a major degradation pathway in eukaryotic systems that allows the removal of large intracellular structures such as entire organelles or protein aggregates, thus contributing to the homeostasis of cells and tissues.

New tricks for the old autophagy protein Atg8

Nature Structural & Molecular Biology, 2021
The autophagy protein Atg8 regulates autophagosome biogenesis and mediates cargo recruitment in different types of selective autophagy, but the molecular mechanisms mediating Atg8 functions during autophagosome biogenesis have been unclear.

In Vitro Assays of Lipidation of Mammalian Atg8 Homologs

Current Protocols in Cell Biology, 2014
AbstractAtg8 modifier in yeast is conjugated to phosphatidylethanolamine via ubiquitylation‐like reactions essential for autophagy. Mammalian Atg8 homologs (Atg8s) including LC3, GABARAP, and GATE‐16, are also ubiquitin‐like modifiers. The carboxyl termini of mammalian Atg8 homologs are cleaved by Atg4B, a cysteine protease, to expose carboxyl terminal

Morphological and cytological analyses of Arabidopsis thaliana seedlings roots growing under conditions mimicking a microgravity has been performed. Moreover, we assessed the process of autophagy induction in cells upon such conditions. Here, we report significant changes in the root development and the enhanced autophagosome formation in epidermal ...

Investigating Structure and Dynamics of Atg8 Family Proteins

2017
Atg8 family members were the first autophagy-related proteins to be investigated in structural detail and continue to be among the best-understood molecules of the pathway. In this review, we will first provide a concise outline of the major methods that are being applied for structural characterization of these proteins and the complexes they are ...
