Effect of developmental changes on pharmacokinetics of drugs used in the treatment of infant acute lymphoblastic leukaemia—A comprehensive review

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
While the event‐free survival (EFS) of children treated for acute lymphoblastic leukaemia (ALL) has improved greatly in the last decades, the EFS for patients diagnosed with ALL before the age of one is still under 50%. This outcome further decreases when infants have a rearrangement in the gene encoding histone‐lysine N‐methyltransferase 2A (KMT2A ...

Assessing transporter‐mediated rifampin–linezolid interaction using physiologically‐based pharmacokinetic modelling

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Abstract Aims The study aims to develop a physiologically‐based pharmacokinetic (PBPK) model to quantitatively evaluate the role of ATP‐binding cassette sub‐family B member 1 (ABCB1) and ATP‐binding cassette super‐family G member 2 (ABCG2) in the drug–drug interaction (DDI) between rifampin and linezolid and to predict the impact of high‐dose rifampin ...

Single-molecule visualization of ATP-induced dynamics of the subunit composition of an ECF transporter complex under turnover conditions

open access: yesNature Communications
Energy-Coupling Factor (ECF) transporters are ATP-binding cassette (ABC) transporters essential for uptake of vitamins and cofactors in prokaryotes. They have been linked to pathogen virulence and are potential targets for antimicrobials.

Efficacy, safety and cost‐effectiveness of CAR‐T therapy

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
CAR T‐cells demonstrate high efficacy in blood cancers, including ALL, MM and DLBCL. Innovations target solid tumours despite challenges such as antigen escape. Combination therapies enhance the delivery and infiltration of CAR T cells. Toxicity, cost and resistance remain major barriers to clinical use.

Adaptive Machine Learning Framework for Optimizing the Affinity Purification of Adeno‐Associated Viral Vectors

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Adeno‐associated viral (AAV) vectors for gene therapy are becoming integral to modern medicine, providing therapeutic options for diseases once deemed incurable. Currently, viral vector purification is a critical bottleneck in the gene therapy industry, impacting product efficacy and safety as well as accessibility and cost to patients ...

The Methods to Overcome ATP-binding Cassette Transporters-mediated Multi-drug Resistance

open access: yesمجله دانشکده پزشکی اصفهان, 2013
Background: Multi-drug resistance (MDR) is a prevalent phenomenon occurred in tumor cells in response to chemotherapy. The main cause of MDR is the efflux of drugs by ATP-binding Cassette (ABC) transporters.
