Antibody repertoire sequencing analysis

open access: yesActa Biochimica et Biophysica Sinica, 2022
High-throughput sequencing for B cell receptor (BCR) repertoire provides useful insights for the adaptive immune system. With the continuous development of the BCR-seq technology, many efforts have been made to develop methods for analyzing the ever ...

Fibrates Inhibit PLTP‐induced M2 Macrophage Infiltration and Increase the Sensitivity of Hepatocellular Carcinoma to ICIs

open access: yesAdvanced Science, EarlyView.
Phospholipid transfer protein(PLTP) plays a critical role in forming a complex with kinase A (AURKA) and P65. This interaction facilitates phosphorylation of P65 at Ser536, leading to the activation of the NF‐κB signaling pathway. Ultimately, this leads to the upregulation of downstream cytokines, including IL‐6, IL‐8, and CSF‐1, which promotes M2 ...

Quantitative Proteomics and CRISPR/Cas9 Editing Reveal UPR‐Mediated Control of Immunoglobulin Homeostasis in Hybridomas

open access: yesAdvanced Science, EarlyView.
BCR sequencing and subclone analysis correlated immunoglobulin (Ig) chain loss in dysfunctional hybridomas with disrupted monoclonal antibody homeostasis. Proteomics‐guided CRISPR/Cas9 editing revealed that the unfolded protein response (UPR) regulates aberrant Ig synthesis.

A Novel Predictor Tool of Biochemical Recurrence after Radical Prostatectomy Based on a Five-MicroRNA Tissue Signature [PDF]

open access: yes, 2019
Within five to ten years after radical prostatectomy (RP), approximately 15-34% of prostate cancer (PCa) patients experience biochemical recurrence (BCR), which is defined as recurrence of serum levels of prostate-specific antigen >0.2 µg/L, indicating ...

Single‐Cell Profiling Across Immune Tissues and Organs Reveals Immunosenescence Signatures in Male Rhesus Monkeys

open access: yesAdvanced Science, EarlyView.
Single‐cell profiling across bone marrow, spleen, mesenteric lymph, and blood in rhesus monkeys reveals organ Immunosenescence. GZMB rises with age, particularly in cytotoxic and terminally exhausted CD8+ T cells, and BHLHE40 emerges as a key transcription factor enriched across multiple CD8+ subsets, regulating pro‐inflammatory and exhaustion‐related ...

Validation of a Drosophila model of wild-type and T315I mutated BCR-ABL1 in chronic myeloid leukemia: an effective platform for treatment screening

open access: yesHaematologica, 2020
Chronic myeloid leukemia (CML) is caused by a balanced chromosomal translocation resulting in the formation of BCR-ABL1 fusion gene encoding a constitutively active BCR-ABL1 tyrosine kinase, which activates multiple signal transduction pathways leading ...

Urinary exosomal microRNA profiling in intermediate-risk prostate cancer

open access: yesScientific Reports, 2021
MicroRNAs (miRNAs) of urine exosomes have emerged as biomarkers for urological cancers, owing to their high stability. MiRNAs have been linked to factors associated with aggressive prostate cancer such as biochemical recurrence (BCR) and metastasis.

A Family History of Lethal Prostate Cancer and Risk of Aggressive Prostate Cancer in Patients Undergoing Radical Prostatectomy. [PDF]

open access: yes, 2015
We investigated whether a family history of lethal prostate cancer (PCa) was associated with high-risk disease or biochemical recurrence in patients undergoing radical prostatectomy. A cohort of radical prostatectomy patients was stratified into men with

A Deep Representation Learning Method for Quantitative Immune Defense Function Evaluation and Its Clinical Applications

open access: yesAdvanced Science, EarlyView.
ImmuDef, a novel algorithm to quantitatively evaluate the anti‐infection immune defense function of an individual based on RNA‐seq data via a variational autoencoder (VAE) model. It is validated on 3200+ samples across four immune states with high accuracy. It can serve as a metric for disease severity and prognosis across pathogenic cohorts.

Chronic myeloid leukemia stem cells are not dependent on Bcr-Abl kinase activity for their survival [PDF]

open access: yes, 2012
Recent evidence suggests CML stem cells are insensitive to kinase inhibitors and responsible for minimal residual disease in treated patients. We investigated whether CML stem cells, in a transgenic mouse model of CML-like disease or derived from ...
