Structural and histone binding ability characterizations of human PWWP domains. [PDF]
The PWWP domain was first identified as a structural motif of 100-130 amino acids in the WHSC1 protein and predicted to be a protein-protein interaction domain. It belongs to the Tudor domain 'Royal Family', which consists of Tudor, chromodomain, MBT and

Isoform-specific involvement of Brpf1 in expansion of adult hematopoietic stem and progenitor cells [PDF]
Abstract Bromodomain-containing proteins are known readers of histone acetylation that regulate chromatin structure and transcription. Although the functions of bromodomain-containing proteins in development, homeostasis, and disease states have been well studied, their role in self-renewal of hematopoietic stem and progenitor cells ...

Molecular Basis of Histone Acetyllysine Recognition by the BRPF1 Bromodomain
The monocytic leukemic zinc‐finger (MOZ) histone acetyltransferase (HAT) was first identified as a fusion partner with the CREB binding protein (CBP) in a chromosomal translocation found in acute myeloid leukemia (AML). Disruption of MOZ HAT activity leads to leukemogenic transformations and oncogenesis.

We report a novel cysteine‐to‐serine surface‐mutation approach to determine co‐crystal structures of KAT6A with small‐molecule inhibitors and compare it with the previously published MYSTcryst approach which uses mutated KAT8 as a surrogate for KAT6A.

Expression atlas of the multivalent epigenetic regulator Brpf1 and its requirement for survival of mouse embryos [PDF]
Bromodomain- and PHD finger-containing protein 1 (BRPF1) is a unique epigenetic regulator that contains multiple structural domains for recognizing different chromatin modifications. In addition, it possesses sequence motifs for forming multiple complexes with three different histone acetyltransferases, MOZ, MORF, and HBO1.

The function of bromodomain‐containing proteins in regulating gene transcription is reviewed here, including the development of chemical probes and targeted protein degradation modalities (e.g., PROTACs). This extends to clinical efforts targeting the BET bromodomain‐containing proteins. Challenges of selectivity, future prospects, and the differential

Unusual backfolded binding poses of BAZ2A bromodomain binders
We identified BAZ2A‐binding compounds assuming a peculiar, almost enclosed, conformation. These molecules pose a basis for the development of potent BAZ2A macrocyclic inhibitors, as performed for other bromodomains.BAZ2A is a large multidomain protein overexpressed in aggressive prostate cancer, where it potentiates migration and invasion of other ...

Conservation Status of Two Italian Local Poultry Breeds: Faraona Camosciata and Oca Padovana
ABSTRACT The present study aimed to assess the conservation status and characterize the phenotypic and genomic diversity of two endangered Italian poultry genetic resources to guide their conservation plans. Blood samples and morphometric data were collected on 50 Faraona Camosciata (FAC, Numida meleagris) and 50 Oca Padovana (OPD, Anser anser) adult ...
