Bumetanide and furosemide

Clinical Pharmacology and Therapeutics, 1983
We assessed the response to and handling of furosemide and bumetanide in 30 experiments with the former and 46 with the latter in normal subjects. Oral doses of furosemide (20, 40, and 80 mg) were used, and subjects received oral doses of 0.5, 1, and 2 mg bumetanide and intravenous doses of 0.5 and 1 mg bumetanide.

Pharmacology of Bumetanide

The Journal of Clinical Pharmacology, 1981
Abstract:The diuretic action of bumetanide was compared with that of furosemide following oral and intravenous administration of equipotent doses in the rat, mouse, cat, rabbit, and dog. The onset of the bumetanide‐induced diuresis was rapid and lasted 4 to 6 hours.

Metabolism of Bumetanide

The Journal of Clinical Pharmacology, 1981
Abstract: The metabolism of bumetanide is reviewed, with emphasis on the relationship between metabolism and diuretic activity. Substantial evidence has been obtained for the thesis that diuretic activity is dependent on the amount of bumetanide which reaches the renal tubule.

In vitro Metabolism of Bumetanide

Pharmaceutical Research, 1984
A new metabolite of the diuretic drug bumetanide, the 4-[(4'-hydroxy)-phenoxy] analog (7), was identified in incubation mixtures of rat liver microsomes. Phenobarbital and clofibrate pretreatment to induce microsomal enzymes changed the relative amounts of the six metabolites formed.

Indomethacin and the response to bumetanide

Clinical Pharmacology and Therapeutics, 1980
Pretreatment of 8 normal subjects with 100 mg indomethacin decreased the response to bumetanide; cumulative 4-hr excretion of sodium due to 1.0 mg of bumetanide was reduced from 276 +/- 22.9 to 202 +/- 20.9 mEq (p less than 0.003). Effects on volume and Cl paralleled those on Na while K excretion was not affected.

Albumin Binding of Bumetanide

Developmental Pharmacology and Therapeutics, 2017
Bumetanide binding to human serum albumin was studied using ultrafiltration. The first stoichiometric binding constant for bumetanide is 6.4 × 10^4 M^-1. Bumetanide competes with bilirubin for human serum albumin binding, having a K(Displ) (displacement constant) of 6.2 × 10^3 M^-1 measured by the peroxidase method.

Bumetanide, a new diuretic

International Urology and Nephrology, 1974
Fifty urological patients with normal renal function, or with either chronic or acute renal failure were treated with Bumetanide infused in saline or a 5% glucose or fructose solution. Thirty-two patients were given Bumetanide exclusively, whereas 18 were given alternating infusions of Bumetanide and Furosemide.
