Rapid and Ultra‐Sensitive SARS‐CoV‐2 Subgenomic RNA Detection Using Single‐Molecule With a Large Transistor‐SiMoT Bioelectronic Platform

open access: yesAdvanced Electronic Materials, EarlyView.
The single‐molecule with a large transistor‐SiMoT is proposed for the detection of the subgenomic RNA of SARS‐CoV‐2, achieving a diagnostic sensitivity of 98.0% and a specificity of 87.8%. The SiMoT technology, currently at TRL‐5, is suitable for point‐of‐care settings and delivers the result to the end user in 30 min.

Major Causes of Rabies Outbreak in Pastoral Areas of the Somali Region and Importance of the One Health Approach

open access: yesAnimal Research and One Health, EarlyView.
Rabies Transmission and Control in the Somali Region. Introduction: Rabies is a fatal zoonotic disease caused by the Lyssavirus, posing a significant threat to both wildlife and public health in the Somali Region of Ethiopia. Key Points: 1. Primary Hosts: Wild carnivores such as foxes, jackals, raccoons, skunks, bats, and honey badgers are the main ...

Development of an HEK293 Suspension Cell Culture Medium, Transient Transfection Optimization Workflow, and Analytics for Batch rAAV Manufacturing

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Recombinant adeno associated virus (rAAV) vectors have become popular delivery vehicles for in vivo gene therapies, but demand for rAAVs continues to outpace supply. Platform processes for rAAV production are being developed by many manufacturers, and transient chemical transfection of human embryonic kidney 293 (HEK293) cells is currently the

Integrating Affinity Chromatography in the Platform Process for Adenovirus Purification

open access: yesBiotechnology and Bioengineering, EarlyView.
Peptide‐functionalized affinity resins with high adenovirus binding capacity, recovery, and contaminant removal afford excellent productivity, and reduced impurities in a three‐step purification process. ABSTRACT Adenoviral vectors (AdVs) are gaining prominence in cancer therapy and vaccine development, posing the need for a modern AdV manufacturing ...

Techno‐Economic Analysis of Membrane‐Based Purification Platforms for AAV Vector Production

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Technologies for large‐scale manufacturing of viral vectors for gene therapies, such as tangential flow filtration and membrane chromatography, are under development. In these early stages of process development, techno‐economic analyses are useful for identifying membrane properties yielding the greatest impact on process performance. In this

Harnessing cell aggregates for enhanced adeno‐associated virus manufacturing: Cultivation strategies and scale‐up considerations

open access: yesBiotechnology Progress, EarlyView.
Abstract The possibility to produce recombinant adeno‐associated virus (rAAV) by adherent HEK293T cells was studied in a stirred tank bioreactor (STR) culture of cell aggregates. A proof‐of‐concept of rAAV production was successfully demonstrated in a process where single cells were first expanded, then cell aggregates were formed by dilution into a ...

Iterative hybrid model based optimization of rAAV production

open access: yesBiotechnology Progress, EarlyView.
Abstract Changes in serotype or genetic payload of recombinant adeno associated virus (rAAVs) gene therapies require adapting the transfection conditions of the upstream HEK293 cultivations. This study adopts an iterative model‐based experiment design approach, where increasing data availability is leveraged to evolve models of different complexity ...

Elasticity Theory and Shape Transitions of Viral Shells

open access: yes, 2005
Recently, continuum elasticity theory has been applied to explain the shape transition of icosahedral viral capsids - single-protein-thick crystalline shells - from spherical to buckled/faceted as their radius increases through a critical value ...

A synthetic platform for developing recombinant adeno‐associated virus type 8 producer cell lines

open access: yesBiotechnology Progress, EarlyView.
Abstract Recombinant adeno‐associated virus (rAAV) is one of the most widely used viral vectors for gene therapy. It is used in very high doses for the treatment of many diseases, making large‐scale production for clinical applications challenging. We have established a synthetic biology‐based platform to construct stable production cell lines, which ...
