Genetic Engineering of Tumor‐Infiltrating Lymphocytes (TIL) via a T‐Editor Platform to Enhance Anti‐Tumor Activity

open access: yesCancer Science, EarlyView.
A rapid and efficient CRISPR‐mediated gene editing platform for TIL engineering identified FAM84B as a novel potential target to enhance antitumor activity and pioneered the use of CBE to generate FAM84B loss‐of‐function TIL with enhanced antitumor activity.

The Role of IGF2BP3‐Mediated Immune Response Activation in Enhancing Immunotherapy of Koumine for Gastric Cancer

open access: yesCancer Science, EarlyView.
Koumine binds IGF2BP3 and promotes its proteasomal degradation, reducing m6A on EED mRNA, accelerating EED decay, lowering H3K27me3, and promoting IRF transcription, thereby restoring MHC‐I and enhancing CD8+ T cell killing. ABSTRACT Resistance to immune checkpoint blockade in gastric cancer often arises from defective antigen presentation due to MHC‐I

SLAMF6 Enhances the Efficacy of Anti‐PD‐L1 Immunotherapy in CRC Through the STAT1/CXCL10 Signaling Axis

open access: yesCancer Science, EarlyView.
SLAMF6 recruits SHP‐2 to activate STAT1, upregulates CXCL10 and PD‐L1, promotes CD8+T cell infiltration, and enhances the efficacy of anti‐PD‐L1 therapy. ABSTRACT As a T‐cell receptor co‐stimulatory protein, signaling lymphocyte activation molecule family 6 (SLAMF6) is a member of the immune cell activating receptor family and is expressed on T cells ...

Spatiotemporal CD8+ T‐Cell Dynamics: Clonal Replacement and Expansion as Determinants of Sustainable Antitumor Response

open access: yesCancer Science, EarlyView.
Durable antitumor immunity depends on continuous clonal replacement: immune checkpoint inhibitors that drive sustainable, polyclonal mobilization of T‐cell clones from tumor‐draining lymph nodes sustain tumor control, whereas limited, oligoclonal mobilization allows pre‐existing clones to become exhausted, leading to immune escape.

Cord Blood‐Derived Fibroblast Activation Protein‐Targeted Chimeric Antigen Receptor Natural Killer Cells Persist in Vivo

open access: yesCancer Science, EarlyView.
We generated cord blood‐derived FAP‐targeted CAR NK cells and evaluated their activity against cancer‐associated fibroblasts. These cells showed antigen‐specific cytotoxicity in vitro and robust persistence in a pleural dissemination model, although in vivo anti‐CAF efficacy was limited, supporting further development of stromal‐targeted cellular ...
